AbstractAccording to the World Health Organization, 45% of deaths among children under five years of age are caused by malnutrition, which impacts more than 224 million children globally. The Barbados Nutrition Study (BNS) is a 50+ year longitudinal study on a Barbadian cohort with histories of moderate to severe protein-energy malnutrition (PEM) limited to the first year of life and a healthy comparison group. We have previously used quantitative electroencephalography (EEG) to highlight differences in brain function duringchildhood(lower alpha1 activity and higher theta, alpha2 and beta activity) between participants who suffered from early PEM and controls. In order to determine whether similar differences between the PEM and control groups persisted intoadulthood, our current study used recordings obtained during a Go-No-Go task in a subsample of the original BNS cohort (N=53) at ages 45-51 years. We found that previously malnourished adults (n=24) had a higher rate of omission errors on the task relative to controls (n=29). Evoked-Related Potentials (ERP) were significantly different in participants with histories of early PEM, who presented with lower N2 amplitudes (p<0.05). These findings are typically associated with impaired conflict monitoring and/or attention deficits and may therefore be linked to the attentional and executive function deficits that have been previously reported in this cohort in childhood and again in middle-adulthood.HighlightsChildhood malnutrition increases risk of brain function alterations. There is a need to investigate the evolution of those outcomes later in life.Adults who suffered childhood malnutrition undertook a Go-No-Go task during EEG.Task performance and N2 amplitude were reduced in malnutrition group (vs control).First evidence of adult brain function alteration following childhood malnutrition.

AbstractAimTo analyse the association between early aEEG and cognitive outcome at early school-age in very preterm infants.MethodsProspective cohort study including infants with gestational age (GA) <32.0 weeks, undergoing continuous aEEG recording during first 4 days of life. Semiquantitative and quantitative (maximum/minimum amplitude) measures were averaged over the recording period. Cognitive outcome was assessed with the Kaufman-Assessment Battery for Children at 5 years of age. Uni- and multivariate logistic regressions were calculated between aEEG parameters and normal cognitive outcome (IQ≥85).ResultsAmong 118 monitored preterm children, 89 were assessed at median(IQR) corrected age of 68.6 months (65.5-71.2) [48% female, median(IQR) GA 29.9(28.2,30.9) weeks, mean(SD) birth weight 1235(363) grams]. Mean(SD) IQ was 97.8(12.7). IQ<85 occurred in 21.3 %, cerebral palsy was found in 3.4%. Despite univariate associations of total maturity scores, cycling subscores, background pattern and minimum aEEG amplitude with normal cognitive outcome none of the associations remained significant after adjustment for confounders. Socioeconomic status was identified as independent predictor of neurodevelopmental outcome.ConclusionIn this cohort of very preterm infants, early short-term aEEG was not predictive of later cognitive outcome. Further research is needed to explore how aEEG could help to inform long-term prognosis in this population group.Key notesPreterm born infants are at high risk for neurodevelopmental impairment.Early amplitude integrated electroencephalography characteristics are univariately associated with cognitive outcome at 5 years of age in preterm born children. However, socioeconomic factors and neonatal morbidity were stronger predictors of long-term neurodevelopmental outcome than early aEEG measures.

AbstractMismarking locations of the fiducials can have a significant influence on the digitized electrode locations and cortical source estimation using high-density EEG. Under-standing and quantifying how uncertainties in the fiducial locations affect the locations of cortical sources is important for interpreting EEG analyses. We systematically shifted fiducial locations to investigate the relationship between variations of fiducial locations and the corresponding estimations of the source locations. We quantified the uncertainty of the dipole locations using the enclosing volume of the dipole locations and the maximum width of the dipole cluster. Shifting fiducial locations 1.5 cm increased the uncertainty of the dipole locations to span a volume >1 cm3 and about 2.5 cm wide. Results suggest that the fiducials need to be digitized accurately within at least 0.5 cm of the absolute actual fiducial location to limit the uncertainty of a dipole location to <1 cm. Additionally, we used random fiducial shift combinations to estimate the effects of combinations of the fiducial shifts on dipole location estimation. This analysis showed that dipole locations were within the bounds of our dipole estimation uncertainty volumes. Based on the outcomes, we suggest marking fiducials carefully before placement of the cap and to use a digitization method with an accuracy of <0.5 cm.

AbstractIn real life, behavior is influenced by dynamically changing contextual factors and is rarely limited to simple tasks and binary choices. For a meaningful interpretation of brain dynamics underlying more natural cognitive processing in active humans, ecologically valid test scenarios are essential. To understand whether brain dynamics in restricted artificial lab settings reflect the neural activity in complex natural environments, we systematically tested the eventrelated P300 in both settings. We developed an integrative approach comprising an initial P300-study in a highly controlled laboratory set-up and a subsequent validation within a realistic driving scenario. Using a simulated dialog with a speech-based input system, increased P300 amplitudes reflected processing of infrequent and incorrect auditory feedback events in both the laboratory setting and the real world setup. Environmental noise and movement-related activity in the car driving scenario led to higher data rejection rates but revealed no effect on signal-to-noise ratio in theta and alpha frequency band or the amplitudes of the event-related P300. Our results demonstrate the possibility to investigate cognitive functions like context updating in highly adverse driving scenarios and encourage the consideration of more realistic task settings in prospective brain imaging approaches.

AbstractA longstanding view holds that information is maintained in working memory (WM) via persistent neural activity that encodes the content of WM. Recent work, however, has challenged the view that all items stored in WM are actively maintained. Instead, “activity-silent” models propose that items can be maintained in WM without the need for persistent neural activity, raising the possibility that only a subset of items – perhaps just a single item – may be actively represented at a given time. While past studies have successfully decoded multiple items stored in WM, these studies cannot rule out an active switching account in which only a single item is actively represented at a time. Here, we directly tested whether multiple representations can be held concurrently in an active state. We tracked spatial representations in WM using alpha-band (8–12 Hz) activity, which encodes spatial positions held in WM. Human observers (male and female) remembered one or two positions over a short delay while we recorded EEG. Using a spatial encoding model, we reconstructed stimulus-specific working memory representations (channel tuning functions, CTFs) from the scalp distribution of alphaband power. Consistent with past work, we found the selectivity of spatial CTFs was lower when two items were stored than when one item was stored. Critically, data-driven simulations revealed that the selectivity of spatial representations in the two-item condition could not be explained by models restricting storage to a single item at a time. Thus, our findings provide robust evidence for the concurrent storage of multiple items in visual working memory.Author SummaryWorking memory (WM) is a mental workspace where we temporarily hold information “online” in pursuit of our current goals. However, recent activity-silent models of WM have challenged the view that all items are held in an “online” state, instead proposing that only a subset of representations in WM – perhaps just one item – are represented by persistent activity at a time. To directly test a single-item model of persistent activity, we used a spatial encoding model to read out the strength of two representations from alpha-band (8–12 Hz) power in the human EEG signal. We provide direct evidence that both locations were maintained concurrently, ruling out the possibility that declines in stimulus-specific activity are due to storing one of two items in an activity-silent state.

ABSTRACTThe outstanding speed of language comprehension necessitates a highly efficient implementation of cognitive-linguistic processes. The domain-general theory of Predictive Coding suggests that our brain solves this problem by continuously forming linguistic predictions about expected upcoming input. The neurophysiological implementation of these predictive linguistic processes, however, is not yet understood. Here, we use EEG (human participants, both sexes) to investigate the existence and nature of online-generated, category-level semantic representations during sentence processing. We conducted two experiments in which some nouns – embedded in a predictive spoken sentence context – were unexpectedly delayed by 1 second. Target nouns were either abstract/concrete (Experiment 1) or animate/inanimate (Experiment 2). We hypothesized that if neural prediction error signals following (temporary) omissions carry specific information about the stimulus, the semantic category of the upcoming target word is encoded in brain activity prior to its presentation. Using time-generalized multivariate pattern analysis, we demonstrate significant decoding of word category from silent periods directly preceding the target word, in both experiments. This provides direct evidence for predictive coding during sentence processing, i.e., that information about a word can be encoded in brain activity before it is perceived. While the same semantic contrast could also be decoded from EEG activity elicited by isolated words (Experiment 1), the identified neural patterns did not generalize to pre-stimulus delay period activity in sentences. Our results not only indicate that the brain processes language predictively, but also demonstrate the nature and sentence-specificity of category-level semantic predictions preactivated during sentence comprehension.STATEMENT OF SIGNIFICANCEThe speed of language comprehension necessitates a highly efficient implementation of cognitive-linguistic processes. Predictive processing has been suggested as a solution to this problem, but the underlying neural mechanisms and linguistic content of such predictions are only poorly understood. Inspired by Predictive Coding theory, we investigate whether the meaning of expected, but not-yet heard words can be decoded from brain activity. Using EEG, we can predict if a word is, e.g., abstract (as opposed to concrete), or animate (vs. inanimate), from brain signals preceding the word itself. This strengthens predictive coding theory as a likely candidate for the principled neural mechanisms underlying online processing of language and indicates that predictive processing applies to highly abstract categories like semantics.

AbstractElectrophysiological methods, i.e., M/EEG provide unique views into brain health. Yet, when building predictive models from brain data, it is often unclear how electrophysiology should be combined with other neuroimaging methods. Information can be redundant, useful common representations of multimodal data may not be obvious and multimodal data collection can be medically contraindicated, which reduces applicability. Here, we propose a multimodal model to robustly combine MEG, MRI and fMRI for prediction. We focus on age prediction as surrogate biomarker in 674 subjects from the Cam-CAN. Strikingly, MEG, fMRI and MRI showed additive effects supporting distinct brain-behavior associations. Moreover, the contribution of MEG was best explained by source-topography of power spectra between 8 and 30 Hz. Finally, we demonstrate that the model maintains benefits of stacking when data is missing. The proposed framework hence enables multimodal learning for a wide range of biomarkers from diverse types of brain signals.

AbstractTranscranial electric stimulation (TES) is an emerging technique, developed to non-invasively modulate brain function. However, the spatiotemporal distribution of the intracranial electric fields induced by TES remains poorly understood. In particular, it is unclear how much current actually reaches the brain, and how it distributes across the brain. Lack of this basic information precludes a firm mechanistic understanding of TES effects. In this study we directly measure the spatial and temporal characteristics of the electric field generated by TES using stereotactic EEG (s-EEG) electrode arrays implanted in cebus monkeys and surgical epilepsy patients. We found a small frequency dependent decrease (10%) in magnitudes of TES induced potentials and negligible phase shifts over space. Electric field strengths were strongest in superficial brain regions with maximum values of about 0.5 mV/mm. Our results provide crucial information for the interpretation of human TES studies and the optimization and design of TES stimulation protocols. In addition, our findings have broad implications concerning electric field propagation in non-invasive recording techniques such as EEG/MEG.

Neuromuscular electrical stimulation (NMES) of the nervous system has been extensively used in neurorehabilitation due to its capacity to engage the muscle fibers, improving muscle tone, and the neural pathways, sending afferent volleys toward the brain. Although different neuroimaging tools suggested the capability of NMES to regulate the excitability of sensorimotor cortex and corticospinal circuits, how the intensity and dose of NMES can neuromodulate the brain oscillatory activity measured with electroencephalography (EEG) is still unknown to date. We quantified the effect of NMES parameters on brain oscillatory activity of 12 healthy participants who underwent stimulation of wrist extensors during rest. Three different NMES intensities were included, two below and one above the individual motor threshold, fixing the stimulation frequency to 35 Hz and the pulse width to 300 μs. Firstly, we efficiently removed stimulation artifacts from the EEG recordings. Secondly, we analyzed the effect of amplitude and dose on the sensorimotor oscillatory activity. On the one hand, we observed a significant NMES intensity-dependent modulation of brain activity, demonstrating the direct effect of afferent receptor recruitment. On the other hand, we described a significant NMES intensity-dependent dose-effect on sensorimotor activity modulation over time, with below-motor-threshold intensities causing cortical inhibition and above-motor-threshold intensities causing cortical facilitation. Our results highlight the relevance of intensity and dose of NMES, and show that these parameters can influence the recruitment of the sensorimotor pathways from the muscle to the brain, which should be carefully considered for the design of novel neuromodulation interventions based on NMES. AbstractThe perceptual consequences of cochlear synaptopathy are presently not well understood as a direct quantification of synaptopathy is not possible in humans. To study its role for human hearing, recent studies have instead correlated changes in basic supra-threshold psychoacoustic tasks with individual differences in subcortical EEG responses, as a proxy measure for synaptopathy. It is not clear whether the reported missing relationships between the psychoacoustic quantities and the EEG are due to the adopted methods, or to a minor role of synaptopathy for sound perception. We address this topic by studying the theoretical relationship between subcortical EEG and psychoacoustic methods for different sensorineural hearing deficits.
