    Former studies suggest that the core features of psychopathy, namely the callous-unemotional (CU) traits are associated with emotional dysfunction characterized by atypical processing of other people's distress. Using a dimensional approach to CU traits, the current study aims to test neural activity during performance of a facial expression recognition task in a community sample of young adults. Forty-one participants (25 females, M age = 25.44, SD = 4.03) completed the Inventory of Callous-Unemotional Traits. Functional magnetic resonance imaging data were collected to measure neural responses to fearful, happy and sad faces as compared with neutral facial expressions. Region-of-interest analyses revealed that during exposure to fearful face expressions, blood oxygenation level-dependent responses were negatively associated with CU traits in the right anterior cingulate gyrus (ACCg), but not in the amygdala. These findings support that CU traits are related to a unique neural response to fearful faces in noncriminal population as well. It also highlights the importance of taking into account other regions outside the amygdala, in particular the ACC, when testing the relation between CU traits and fear response. © 2017 The Authors

    Here we show a method of directing the edges of the connectomes, prepared from diffusion tensor imaging (DTI) datasets from the human brain. Before the present work, no high-definition directed braingraphs (or connectomes) were published, because the tractography methods in use are not capable of assigning directions to the neural tracts discovered. Previous work on the functional connectomes applied low-resolution functional MRI-detected statistical causality for the assignment of directions of connectomes of typically several dozens of vertices. Our method is based on the phenomenon of the "Consensus Connectome Dynamics" (CCD), described earlier by our research group. In this contribution, we apply the method to the 423 braingraphs, each with 1015 vertices, computed from the public release of the Human Connectome Project, and we also made the directed connectomes publicly available at the site \url{http://braingraph.org}. We also show the robustness of our edge directing method in four independently chosen connectome datasets: we have found that 86\% of the edges, which were present in all four datasets, get the very same directions in all datasets; therefore the direction method is robust, it does not depend on the particular choice of the dataset. We think that our present contribution opens up new possibilities in the analysis of the high-definition human connectome: from now on we can work with a robust assignment of directions of the connections of the human brain.

    Transforming growth factor-βs (TGF-βs) regulate cellular proliferation, differentiation, and survival. TGF-βs bind to type I (TGF-βRI) and II receptors (TGF-βRII), which are transmembrane kinase receptors, and an accessory type III receptor (TGF-βRIII). TGF-β may utilize another type I receptor, activin-like kinase receptor (Alk1). TGF-β is neuroprotective in the middle cerebral artery occlusion (MCAO) model of stroke. Recently, we reported the expression pattern of TGF-β1-3 after MCAO. To establish how TGF-βs exert their actions following MCAO, the present study describes the induction of TGF-βRI, RII, RIII and Alk1 at 24 h, 72 h and 1 mo after transient 1 h MCAO as well as following 24 h permanent MCAO using in situ hybridization histochemistry. In intact brain, only TGF-βRI had significant expression: neurons in cortical layer IV contained TGF-βRI. At 24 h after the occlusion, no TGF-β receptors showed induction. At 72 h following MCAO, all four types of TGF-β receptors were induced in the infarct area, while TGF-βRI and RII also appeared in the penumbra. Most cells with elevated TGF-βRI mRNA levels were microglia. TGF-βRII co-localized with both microglial and endothelial markers while TGF-βRIII and Alk1 were present predominantly in endothels. All four TGF-β receptors were induced within the lesion 1 mo after the occlusion. In particular, TGF-βRIII was further induced as compared to 72 h after MCAO. At this time point, TGF-βRIII signal was predominantly not associated with blood vessels suggesting its microglial location. These data suggest that TGF-β receptors are induced after MCAO in a timely and spatially regulated fashion. TGF-β receptor expression is preceded by increased TGF-β expression. TGF-βRI and RII are likely to be co-expressed in microglial cells while Alk1, TGF-βRII, and RIII in endothels within the infarct where TGF-β1 may be their ligand. At later time points, TGF-βRIII may also appear in glial cells to potentially affect signal transduction via TGF-βRI and RII.

    AbstractWe published a study recently testing the link between brain size and behavioural plasticity using brain size selected guppy (Poecilia reticulata) lines (2019, Journal of Evolutionary Biology, 32, 218‐226). Only large‐brained fish showed habituation to a new, but actually harmless environment perceived as risky, by increasing movement activity over the 20‐day observation period. We concluded that “Our results suggest that brain size likely explains some of the variation in behavioural plasticity found at the intraspecific level”. In a commentary published in the same journal, Haave‐Audet et al. challenged the main message of our study, stating that (a) relative brain size is not a suitable proxy for cognitive ability and (b) habituation measured by us is likely not adaptive and costly. In our response, we first show that a decade's work has proven repeatedly that relative brain size is indeed positively linked to cognitive performance in our model system. Second, we discuss how switching from stressed to unstressed behaviour in stressful situations without real risk is likely adaptive. Finally, we point out that the main cost of behavioural plasticity in our case is the development and maintenance of the neural system needed for information processing, and not the expression of plasticity. We hope that our discussion with Haave‐Audet et al. helps clarifying some central issues in this emerging research field.

    Abstract. We present the analysis aimed at the estimation of flood risks of Tisza River in Hungary on the basis of daily river discharge data registered in the last 100 years. The deseasonalised series has skewed and leptokurtic distribution and various methods suggest that it possesses substantial long memory. This motivates the attempt to fit a fractional ARIMA model with non-Gaussian innovations as a first step. Synthetic streamflow series can then be generated from the bootstrapped innovations. However, there remains a significant difference between the empirical and the synthetic density functions as well as the quantiles. This brings attention to the fact that the innovations are not independent, both their squares and absolute values are autocorrelated. Furthermore, the innovations display non-seasonal periods of high and low variances. This behaviour is characteristic to generalised autoregressive conditional heteroscedastic (GARCH) models. However, when innovations are simulated as GARCH processes, the quantiles and extremes of the discharge series are heavily overestimated. Therefore we suggest to fit a smooth transition GARCH-process to the innovations. In a standard GARCH model the dependence of the variance on the lagged innovation is quadratic whereas in our proposed model it is a bounded function. While preserving long memory and eliminating the correlation from both the generating noise and from its square, the new model is superior to the previously mentioned ones in approximating the probability density, the high quantiles and the extremal behaviour of the empirical river flows.

    We estimate the well-distribution measure and correlation of order 2 of the binary sequence $E_N=\{e_1,\ldots,e_N\}$ defined by $e_n=+1$ if $0\leqslant\{n^c\} \lt {1}/{2}$ and $e_n=-1$ if ${1}/{2}\leqslant\{n^c\} \lt 1$, where $c$ is a real, non-integral number greater than $1$ and $\{x\}$ denotes the fractional part of $x$. We also prove an upper bound for the well-distribution measure of an arbitrary binary sequence in terms of its generating function and show that there exists no upper bound of this type for the correlation. The proof is based on the Erdős-Turán inequality, which we establish with an improved constant.

    Abstract Zearalenone (ZEA) is a mycotoxin produced by Fusarium species, detectable in various cereals and processed food products worldwide. ZEA displays a significant estrogenic activity, thus its main health risk is the interference with sexual maturation and reproduction processes. However, in addition to being key hormonal regulators of reproductive function, estrogenic compounds have a widespread role in brain, as neurotrophic and neuroprotective factors, and they may influence the activity of several brain areas not directly linked to reproduction, as well. Therefore, in the present study, acute effects of ZEA were studied on certain neuronal functions in rats. Experiments were performed on rat brain slices or live rats. Slices were incubated in ZEA-containing (10–100 μM) solution for 30 min. Electrically evoked and spontaneous field potentials were studied in the neocortex and in the hippocampus. At higher concentrations, ZEA incubation of the slices altered excitability and the pattern of epileptiform activity in neocortex and inhibited the development of LTP in hippocampus. For the verification of these in vitro results, in vivo electrophysiological and immunohistochemical investigations were also performed. ZEA was administered systemically (5 mg/kg, i.p.) to male rats and somatosensory evoked potentials and neuronal activation studied by c-fos expression were analyzed. No neuronal activation could be demonstrated in the hippocampus within 2 h of the injection. In the somatosensory cortex, ZEA did not change in vivo evoked potential parameters, but the activation of a small neuronal population could be demonstrated with the c-fos technique in this brain area. This result could be associated with the ZEA-induced alteration of epileptiform activity observed in vitro. Altogether, the toxin altered the excitability and plasticity of neuronal networks after direct treatment in slices, but the effects were less prominent on the given brain areas after systemic treatment in vivo. A probable explanation for the partial lack of in vivo effects may be that after a single injection, ZEA did not cross the blood-brain barrier at sufficient rate to allow the build-up of comparable concentrations in the investigated brain areas. However, in case of compromised blood-brain barrier functions or long-term repeated exposure, alterations in cortical and hippocampal functions cannot be ruled out.

    Abstract. We present a near global statistics on the correlation properties of daily temperature records. Data from terrestrial meteorological stations in the Global Daily Climatology Network are analyzed by means of detrended fluctuation analysis. Long-range temporal correlations extending up to several years are detected for each station. In order to reveal nonlinearity, we evaluated the magnitude of daily temperature changes (volatility) by the same method. The results clearly indicate the presence of nonlinearities in temperature time series, furthemore the geographic distribution of correlation exponents exhibits well defined clustering.

    Human trypsin 4 is an unconventional serine protease that possesses an arginine at position 193 in place of the highly conserved glycine. Although this single amino acid substitution does not affect steady-state activity on small synthetic substrates, it has dramatic effects on zymogen activation, interaction with canonical inhibitors, and substrate specificity toward macromolecular substrates. To study the effect of a non-glycine residue at position 193 on the mechanism of the individual enzymatic reaction steps, we expressed wild type human trypsin 4 and its R193G mutant. 4-Methylumbelliferyl 4-guanidinobenzoate has been chosen as a substrate analogue, where deacylation is rate-limiting, and transient kinetic methods were used to monitor the reactions. This experimental system allows for the separation of the individual reaction steps during substrate hydrolysis and the determination of their rate constants dependably. We suggest a refined model for the reaction mechanism, in which acylation is preceded by the reversible formation of the first tetrahedral intermediate. Furthermore, the thermodynamics of these steps were also investigated. The formation of the first tetrahedral intermediate is highly exothermic and accompanied by a large entropy decrease for the wild type enzyme, whereas the signs of the enthalpy and entropy changes are opposite and smaller for the R193G mutant. This difference in the energetic profiles indicates much more extended structural and/or dynamic rearrangements in the equilibrium step of the first tetrahedral intermediate formation in wild type human trypsin 4 than in the R193G mutant enzyme, which may contribute to the biological function of this protease.

    Human listeners can focus on one speech stream out of several concurrent ones. The present study aimed to assess the whole-brain functional networks underlying a) the process of focusing attention on a single speech stream vs. dividing attention between two streams and 2) speech processing on different time-scales and depth. Two spoken narratives were presented simultaneously while listeners were instructed to a) track and memorize the contents of a speech stream and b) detect the presence of numerals or syntactic violations in the same ("focused attended condition") or in the parallel stream ("divided attended condition"). Speech content tracking was found to be associated with stronger connectivity in lower frequency bands (delta band- 0,5-4 Hz), whereas the detection tasks were linked with networks operating in the faster alpha (8-10 Hz) and beta (13-30 Hz) bands. These results suggest that the oscillation frequencies of the dominant brain networks during speech processing may be related to the duration of the time window within which information is integrated. We also found that focusing attention on a single speaker compared to dividing attention between two concurrent speakers was predominantly associated with connections involving the frontal cortices in the delta (0.5-4 Hz), alpha (8-10 Hz), and beta bands (13-30 Hz), whereas dividing attention between two parallel speech streams was linked with stronger connectivity involving the parietal cortices in the delta and beta frequency bands. Overall, connections strengthened by focused attention may reflect control over information selection, whereas connections strengthened by divided attention may reflect the need for maintaining two streams in parallel and the related control processes necessary for performing the tasks.

    Former studies suggest that the core features of psychopathy, namely the callous-unemotional (CU) traits are associated with emotional dysfunction characterized by atypical processing of other people's distress. Using a dimensional approach to CU traits, the current study aims to test neural activity during performance of a facial expression recognition task in a community sample of young adults. Forty-one participants (25 females, M age = 25.44, SD = 4.03) completed the Inventory of Callous-Unemotional Traits. Functional magnetic resonance imaging data were collected to measure neural responses to fearful, happy and sad faces as compared with neutral facial expressions. Region-of-interest analyses revealed that during exposure to fearful face expressions, blood oxygenation level-dependent responses were negatively associated with CU traits in the right anterior cingulate gyrus (ACCg), but not in the amygdala. These findings support that CU traits are related to a unique neural response to fearful faces in noncriminal population as well. It also highlights the importance of taking into account other regions outside the amygdala, in particular the ACC, when testing the relation between CU traits and fear response. © 2017 The Authors
