    Academic centers utilize sequential clinical and neuroimaging assessments, including morphometric ratios, to obtain an unequivocal diagnosis of the non-synucleinopathic forms of Parkinsonism, such as progressive supranuclear palsy (PSP), however, a 1–2 year follow-up is required. The on-going long-lasting trials using anti-tau antibodies for PSP patients might therefore be biased by the incorrect enrollment of Parkinson’s disease (PD) patients manifesting early axial signs. This perspective study aimed at achieving two major goals: first, to summarize the established biomarker candidates found in cerebrospinal fluid (CSF) in probable PSP patients, including low p-tau and altered neurofilaments. Second, we share our recent data, from CSF samples of well-selected PSP subjects, attributable to both main variants (and revisited in light of MDS criteria), who were followed for 1 year before and 2 years after lumbar puncture. We found a significantly high level of noradrenaline (NE) in these patients, similar to controls, when compared to PD patients. In contrast, CSF samples, in PD, showed a significant reduction in CSF NE and its major metabolite, which confirmed that PD is a multi-system disease involving several endogenous pathways. The NE axis impairments were prominent in PSP featuring worse NPI. It might represent a counterpart to the early and peculiar psycho-pathological profiles that are observed in tauopathies. In conclusion, we highlight that CSF biomarkers, which are easy to collect, can provide rapid insights as diagnostic tools. Early alterations in endogenous NE machinery in atypical Parkinsonism may represent a specific risk trait in forms characterized by a worse prognosis.

    We measure photometric redshifts and spectral types for galaxies in the COSMOS survey. We use template fitting technique combined with luminosity function priors and with the option to simultaneously estimate dust extinction (i.e. E(B-V)) for each galaxy.Our estimated redshifts are accurate to i<25 and z~1.2. Using simulations with sampling and noise characteristics similar to those in COSMOS, the accuracy and reliability is estimated for the photometric redshifts as a function of the magnitude limits of the sample, S/N ratios and the number of bands used. From the simulations we find that the ratio of derived 95% confidence interval in the redshift probability distribution to the estimated photometric redshift (D95) can be used to identify and exclude the catastrophic failures in the photometric redshift estimates. We compare the derived redshifts with high-reliability spectroscopic redshifts for a sample of 868 normal galaxies with z < 1.2 from zCOSMOS. Considering different scenarios, depending on using prior, no prior and/or extinction, we compare the photometric and spectroscopic redshifts for this sample. This corresponds to an rms scatter of 0.031, with a small number of outliers (<2.5%). We also find good agreement (rms=0.10) between photometric and spectroscopic redshifts for Type II AGNs. We compare results from our photometric redshift procedure with three other independent codes and find them in excellent agreement. We show preliminary results, based on photometric redshifts for the entire COSMOS sample (to i < 25 mag.). 38 pages; 14 Figures; 7 Tables. Accepted for Publication in ApJS. COSMOS Special Issue

    Mercury (Hg), which is a non-essential element, is considered a highly toxic pollutant for biological systems even when present at trace levels. Elevated Hg exposure with the growing release of atmospheric pollutant Hg and rising accumulations of mono-methylmercury (highly neurotoxic) in seafood products have increased its toxic potential for humans. This review aims to highlight the potential relationship between Hg exposure and Alzheimer's disease (AD), based on the existing literature in the field. Recent reports have hypothesized that Hg exposure could increase the potential risk of developing AD. Also, AD is known as a complex neurological disorder with increased amounts of both extracellular neuritic plaques and intracellular neurofibrillary tangles, which may also be related to lifestyle and genetic variables. Research reports on AD and relationships between Hg and AD indicate that neurotransmitters such as serotonin, acetylcholine, dopamine, norepinephrine, and glutamate are dysregulated in patients with AD. Many researchers have suggested that AD patients should be evaluated for Hg exposure and toxicity. Some authors suggest further exploration of the Hg concentrations in AD patients. Dysfunctional signaling pathways in AD and Hg exposure appear to be interlinked with some driving factors such as arachidonic acid, homocysteine, dehydroepiandrosterone (DHEA) sulfate, hydrogen peroxide, glucosamine glycans, glutathione, acetyl-L carnitine, melatonin, and HDL. This evidence suggests the need for a better understanding of the relationship between AD and Hg exposure, and potential mechanisms underlying the effects of Hg exposure on regional brain functions. Also, further studies evaluating brain functions are needed to explore the long-term effects of subclinical and untreated Hg toxicity on the brain function of AD patients.

    Objective: Central cholinergic circuits of human brain can be tested non-invasively by coupling peripheral nerve stimulation with transcranial magnetic stimulation of motor cortex. This test, named short latency afferent inhibition (SAI) has been shown in healthy subjects to be sensitive to the blockage of muscarinic acetylcholine receptors and it is impaired in Alzheimer disease (AD) patients, a cholinergic form of dementia, while it is normal in non-cholinergic forms of dementia such as fronto-temporal dementia. The objective of present study was to evaluate central cholinergic circuits in patients with Vascular Dementia (VaD). Methods: We evaluated SAI in a group of patients with VaD and compared the data with those from a group of AD patients and a control group of age-matched healthy individuals. Results: Mean SAI was normal in VaD patients while it was significantly reduced in AD patients. The analysis of individual data showed abnormal SAI in 75% of AD and in only 25% of VaD. Conclusions: SAI is normal in most of VaD patients in contrast with AD patients. This test might be used for the functional evaluation of central cholinergic circuits in VaD patients. Significance: SAI testing may represent a useful additional tool for the evaluation of patients with VaD however, further studies are required in order to evaluate whether this method can be used for the differential diagnosis between pure VaD and different forms of dementia.

    The 2-[14C]deoxyglucose method was used to examine the effects of acute intravenous administration of methamphetamine (0.5-2.5 mg/kg) on rates of local cerebral glucose utilization in freely-moving rats. These effects were correlated with the effects of methamphetamine on locomotor activity assessed simultaneously in the same animals. Methamphetamine administration resulted in widespread dose-dependent increases in glucose utilization within structures of the extrapyramidal motor system. Rates of glucose utilization were positively correlated with locomotor activity in the globus pallidus, substantia nigra reticulata, entopeduncular nucleus, subthalamic nucleus, and the lateral cerebellar cortex. In contrast, within the limbic system alterations in metabolic activity were smaller and more selective. Glucose utilization was increased in the nucleus accumbens at all doses tested, but alterations in glucose utilization in the ventral tegmental area, amygdala, and anterior cingulate were observed only at the highest doses of methamphetamine tested. Significant increases in rates of glucose metabolism were also found in the substantia nigra compacta and in the median and dorsal raphe nuclei. Dopamine and serotonin are depleted in these regions, as well as in the ventral tegmental area where glucose utilization was also increased, following chronic treatment with high doses of methamphetamine. These changes in glucose utilization may be indicative of disturbances in the biochemical processes involved in the neurotoxic effects of methamphetamine.

    Research in genetics of epilepsy represents an area of great interest both for clinical purposes and for understanding the basic mechanisms of epilepsy. Most mutations in epilepsies without structural brain abnormalities have been identified in ion channel genes, but an increasing number of genes involved in a diversity of functional and developmental processes are being recognized through whole exome or genome sequencing. Targeted molecular diagnosis is now available for different forms of epilepsy. The identification of epileptogenic mutations in patients before epilepsy onset and the possibility of developing therapeutic strategies tested in experimental models may facilitate experimental approaches that prevent epilepsy or decrease its severity. Functional analysis is essential for better understanding pathogenic mechanisms and gene interactions. In vitro experimental systems are either cells that usually do not express the protein of interest or neurons in primary cultures. In vivo/ex vivo systems are organisms or preparations obtained from them (e.g., brain slices), which should better model the complexity of brain circuits and actual pathophysiological conditions. Neurons differentiated from induced pluripotent stem cells generated from the skin fibroblasts of patients have recently allowed the study of mutations in human neurons having the genetic background of a given patient. However, there is remarkable complexity underlying epileptogenesis in the clinical dimension, as reflected by the fact that experimental models have not provided yet results having clinical translation and that, with a few exceptions concerning rare conditions, no new curative treatment has emerged from any genetic finding in epilepsy.

    Ces dernieres annees, des etudes ont ete faites en Sardaigne pour ameliorer le traitement de la varroose par l'Apistan® dans des colonies renfermant du couvain opercule d'ouvrieres. Notre experimentation, visant principalement a verifier la persistance de la matiere active dans les lanieres Apistan®, a ete realisee en septembre 1994 dans une region ayant une faible densite de colonies d'abeilles et a porte sur 12 colonies d'Apis mellifera ligustica dans des ruches Dadant-Blatt. L'efficacite du traitement a ete estimee en comptant les acariens tombes sur une feuille de plastique blanche enduite de vaseline et placee sur le fond de chaque ruche avant (2 semaines), pendant (10 semaines) et apres (4 semaines) le traitement. Avant et apres le traitement, nous avons aussi estime: i) la quantite d'abeilles adultes et le couvain opercule, utilisant pour cela un sixieme (188 cm 2 ) d'un cadre Dadant-Blatt comme unite de mesure (Marchetti, 1985), ii) l'infestation des abeilles adultes en prelevant 500 abeilles/ruche reparties sur trois cadres (Ritter et Ruttner, 1980) et iii) l'infestation du couvain opercule d'ouvrieres en inspectant 300 cellules/ruche reparties sur trois cadres et ouvertes de maniere croisee pour surmonter le probleme de la distribution irreguliere de l'acarien dans le nid a couvain (Pappas et Thrasyvoulou, 1988). Pour evaluer la teneur en fluvalinate des lanieres, de petits disques (5 mm de diametre et 20 mg) ont ete preleves chaque semaine dans chaque laniere a l'aide d'un poincon. La teneur en fluvalinate est restee pratiquement constante tout au long des 10 semaines de traitement. On n'a observe aucune difference significative entre les concentrations initiale (11,5 ± 0,6 %) et finale (10,7 ± 0,3 %) en fluvalinate (tableau I), ce qui indique que le prelevement par les abeilles est insignifiant et que le dosage en matiere active de la laniere est plus que suffisant pour controler la varroose. L'efficacite au champ a ete > 99 %. Au cours du traitement, on a denombre 3 615 ± 1 187 acariens/ruche tombes sur le fond. La mortalite des acariens est survenue en majorite (> 97 %) au cours des 4 premieres semaines. L'examen apres le traitement a montre une mortalite naturelle supplementaire de 0,6 ± 0,2 % acarien/ruche/jour comparee a une mortalite naturelle avant traitement de 11,5 ± 2,9 % acarien/ruche/jour. Les infestations moyennes des nymphes et des adultes sont passees de 15,7 ± 7,3 % a zero et de 14,2 ± 7,3 % a zero, respectivement. Sur la base de ces resultats, la reutilisation des lanieres Apistan® serait justifiee (Pechhacker, 1991). Neanmoins, le maintien des lanieres dans la ruche doit etre limite au minimum necessaire (4 semaines) afin de reduire les risques toxicologiques et biologiques.

    Rosmarinic acid (RA), which has multiple bioactive properties, might be a useful agent for protecting central nervous system against age related alterations. In this context, the purpose of the present study was to investigate possible protective effects of RA on mismatch negativity (MMN) component of auditory event-related potentials (AERPs) as an indicator of auditory discrimination and echoic memory in the ovariectomized (OVX) rats injected with D-galactose combined with neurochemical and histological analyses. Ninety female Wistar rats were randomly divided into six groups: sham control (5); RA-treated (R); OVX (O); OVX + RA-treated (OR); OVX+ D-galactose-treated (OD); OVX+ D-galactose + RA treated (ODR). Eight weeks later, MMN responses were recorded using the oddball condition. An amplitude reduction of some components of AERPs was observed due to ovariectomy with or without D-galactose administiration and these reduction patterns were diverse for different electrode locations. MMN amplitudes were significantly lower over temporal and right frontal locations in the o and OD groups versus the S and R groups, which was accompanied by increased thiobarbituric acid reactive substances (TBARS) and hydroxy-2-nonenal (4-HNE) levels. RA treatment significantly increased AERP/MMN amplitudes and lowered the TBARS/4-HNE levels in the OR and ODR groups versus the 0 and OD groups, respectively. Our findings support the potential benefit of RA in the prevention of auditory distortion related to the estrogen deficiency and D-galactose administration at least partly by antioxidant actions.     Background. Magnetic Resonance (MR) diffusion tensor imaging (DTI) is able to quantify in vivo tissue microstructure properties and to detect disease related pathology of the central nervous system. We applied 6 methods for SNR computation in 26 DTI sequences with different parameters using 4 healthy volunteers (HV). We choosed two DTI sequences for their high SNR, they differed by voxel size and b-value. Subsequently, the two selected sequences were acquired from 30 multiple sclerosis (MS) patients with different disability and lesion load and 18 age matched HV. We observed high concordance between mean diffusivity (MD) and fractional anysotropy (FA), nonetheless the DTI sequence with smaller voxel size displayed a better correlation with disease progression, despite a slightly lower SNR. The reliability of corpus callosum (CC) fiber tracking with the chosen DTI sequences was also tested.Conclusion. The sensitivity of DTI-derived indices to MS-related tissue abnormalities indicates that the optimized sequence may be a powerful tool in studies aimed at monitoring the disease course and severity.

    A morphosyntactic agreement violation during reading elicits a well-documented biphasic ERP pattern (LAN+P600). The cognitive variables that affect both the amplitude of the two components and the topography of the anterior negativity are still debated. We studied the ERP correlates of the violation of a specific agreement feature based on the phonology of the critical word. This was compared with the violation of a lexical feature, namely grammatical gender. These two features are different both in the level of representation involved in the agreement computation and in terms of their role in establishing structural relations with possible following constituents. The ERP pattern elicited by the two agreement violations showed interesting dissociations. The LAN was distributed ventrally for both types of violation, but showed a central extension for the gender violation. The P600 showed an amplitude modulation: this component was larger for phonotactic violations in its late time window (700-900 ms). The former result is indicative of a difference in the brain structures recruited for the processing of violations at different levels of representation. The P600 effect is interpreted assuming a hierarchical relation among features that forces a deeper reanalysis of the violation involving a word form property. Finally the two features elicit distinct end-of-sentence wrap-up effects, consistent with the different roles they play in the processing of the whole sentence.

    Academic centers utilize sequential clinical and neuroimaging assessments, including morphometric ratios, to obtain an unequivocal diagnosis of the non-synucleinopathic forms of Parkinsonism, such as progressive supranuclear palsy (PSP), however, a 1–2 year follow-up is required. The on-going long-lasting trials using anti-tau antibodies for PSP patients might therefore be biased by the incorrect enrollment of Parkinson’s disease (PD) patients manifesting early axial signs. This perspective study aimed at achieving two major goals: first, to summarize the established biomarker candidates found in cerebrospinal fluid (CSF) in probable PSP patients, including low p-tau and altered neurofilaments. Second, we share our recent data, from CSF samples of well-selected PSP subjects, attributable to both main variants (and revisited in light of MDS criteria), who were followed for 1 year before and 2 years after lumbar puncture. We found a significantly high level of noradrenaline (NE) in these patients, similar to controls, when compared to PD patients. In contrast, CSF samples, in PD, showed a significant reduction in CSF NE and its major metabolite, which confirmed that PD is a multi-system disease involving several endogenous pathways. The NE axis impairments were prominent in PSP featuring worse NPI. It might represent a counterpart to the early and peculiar psycho-pathological profiles that are observed in tauopathies. In conclusion, we highlight that CSF biomarkers, which are easy to collect, can provide rapid insights as diagnostic tools. Early alterations in endogenous NE machinery in atypical Parkinsonism may represent a specific risk trait in forms characterized by a worse prognosis.

    We measure photometric redshifts and spectral types for galaxies in the COSMOS survey. We use template fitting technique combined with luminosity function priors and with the option to simultaneously estimate dust extinction (i.e. E(B-V)) for each galaxy.Our estimated redshifts are accurate to i<25 and z~1.2. Using simulations with sampling and noise characteristics similar to those in COSMOS, the accuracy and reliability is estimated for the photometric redshifts as a function of the magnitude limits of the sample, S/N ratios and the number of bands used. From the simulations we find that the ratio of derived 95% confidence interval in the redshift probability distribution to the estimated photometric redshift (D95) can be used to identify and exclude the catastrophic failures in the photometric redshift estimates. We compare the derived redshifts with high-reliability spectroscopic redshifts for a sample of 868 normal galaxies with z < 1.2 from zCOSMOS. Considering different scenarios, depending on using prior, no prior and/or extinction, we compare the photometric and spectroscopic redshifts for this sample. This corresponds to an rms scatter of 0.031, with a small number of outliers (<2.5%). We also find good agreement (rms=0.10) between photometric and spectroscopic redshifts for Type II AGNs. We compare results from our photometric redshift procedure with three other independent codes and find them in excellent agreement. We show preliminary results, based on photometric redshifts for the entire COSMOS sample (to i < 25 mag.). 38 pages; 14 Figures; 7 Tables. Accepted for Publication in ApJS. COSMOS Special Issue
