    One of the most severe types of cancer caused by the uncontrollable proliferation of brain cells inside the skull is brain tumors. Hence, a fast and accurate tumor detection method is critical for the patient’s health. Many automated artificial intelligence (AI) methods have recently been developed to diagnose tumors. These approaches, however, result in poor performance; hence, there is a need for an efficient technique to perform precise diagnoses. This paper suggests a novel approach for brain tumor detection via an ensemble of deep and hand-crafted feature vectors (FV). The novel FV is an ensemble of hand-crafted features based on the GLCM (gray level co-occurrence matrix) and in-depth features based on VGG16. The novel FV contains robust features compared to independent vectors, which improve the suggested method’s discriminating capabilities. The proposed FV is then classified using SVM or support vector machines and the k-nearest neighbor classifier (KNN). The framework achieved the highest accuracy of 99% on the ensemble FV. The results indicate the reliability and efficacy of the proposed methodology; hence, radiologists can use it to detect brain tumors through MRI (magnetic resonance imaging). The results show the robustness of the proposed method and can be deployed in the real environment to detect brain tumors from MRI images accurately. In addition, the performance of our model was validated via cross-tabulated data.

    AbstractGuilt, a self-conscious emotion, includes self-focused role taking and also correlates with other-oriented role-taking. Excess guilt proneness might be relevant to obsessive compulsive disorders. The white matter (WM) neural correlates of the degree of guilt have not yet been determined. We hypothesized that the WM structures involved in feelings of guilt are associated with social and moral cognition (inferior parietal lobule [IPL], prefrontal cortex [PFC], and cingulate), and aimed to visualize this using diffusion MRI. We investigated the association between regional WM structures (WM volume, and fractional anisotropy, and mean diffusivity [MD]), and feelings of guilt in 1196 healthy, young students using MRI and the Guilty Feeling Scale, which comprises interpersonal situation (IPS; guilt from hurting friends) and rule-breaking situation (RBS; deontological guilt) scores. The primary novel finding presented here is that MD in the right somatosensory and motor cortices from arm to hand were positively correlated with RBS scores. Further, consistent with our hypothesis, RBS scores were positively correlated with MD in the same regions. These results would be predicted by the Macbeth effect, an obsession with dirt leading to hand-washing rituals resulting from guilt, made famous by the Shakespearian character Lady Macbeth. “What, will these hands ne’er be clean?” William Shakespeare (Shakespeare, 1606) Macbeth.

    Although exact causes of Parkinson disease (PD) remain enigmatic, mitochondrial dysfunction is increasingly appreciated as a key determinant of dopaminergic neuron susceptibility in both familial and sporadic PD. Two genes associated with recessive, early-onset PD encode the ubiquitin (Ub) kinase PINK1 and the E3 Ub ligase PRKN/PARK2/Parkin, which together orchestrate a protective mitochondrial quality control (mitoQC) pathway. Upon stress, both enzymes cooperatively identify and decorate damaged mitochondria with phosphorylated poly-Ub (p-S65-Ub) chains. This specific label is subsequently recognized by autophagy receptors that further facilitate mitochondrial degradation in lysosomes (mitophagy). Here, we analyzed human post-mortem brain specimens and identified distinct pools of p-S65-Ub-positive structures that partially colocalized with markers of mitochondria, autophagy, lysosomes and/or granulovacuolar degeneration bodies. We further quantified levels and distribution of the ‘mitophagy tag’ in 2 large cohorts of brain samples from normal aging and Lewy body disease (LBD) cases using unbiased digital pathology. Somatic p-S65-Ub structures independently increased with age and disease in distinct brain regions and enhanced levels in LBD brain were age- and Braak tangle stage-dependent. Additionally, we observed significant correlations of p-S65-Ub with LBs and neurofibrillary tangle levels in disease. The degree of co-existing p-S65-Ub signals and pathological PD hallmarks increased in the pre-mature stage, but decreased in the late stage of LB or tangle aggregation. Altogether, our study provides further evidence for a potential pathogenic overlap among different forms of PD and suggests that p-S65-Ub can serve as a biomarker for mitochondrial damage in aging and disease. 11,059 4,168 1418 1404 SCIE Q1

    Antiparasitic agents against Taenia solium cysticercosis have been in use since 1979, although its use has been questioned on the basis that cysts would die naturally and thus treatment-induced inflammation is unnecessary. In addition, isolated reports have also questioned whether neurocysticercosis (NCC) is a cause of epilepsy. After more than three and a half decades, a large body of evidence is available. Little if any doubt exists about NCC as a cause of seizures - NCC is consistently associated with seizures when appropriate groups are compared, and in a large subset of cases, seizure semiology correlates with the anatomical location of lesions. Cyst degeneration and the subsequent inflammatory reaction increase seizure expression, although patients with non-inflamed cysts may have seizures, as do patients with long-standing, not inflamed calcified scars. Assessment of the evidence on cysticidal efficacy, safety, and the impact of cyst destruction in decreasing seizures leads to the conclusion that the benefits of antiparasitic treatment in parenchymal brain cysticercosis clearly outweigh the risks, and have provided substantive evidence of the role of NCC as a cause of seizures and epilepsy. Antiparasitic therapy should be considered a primary option in the management of patients with live or degenerating brain NCC cysts.     We recently reported that exercise increases resilience to stress in young female mice. Underlying mechanisms include an interaction of the ovarian hormone estradiol (E2) with insulin-like growth factor I (IGF-I), and an increase in the hippocampal levels of the latter. Since changes in mood regulation during aging may contribute to increasing incidence of affective disorders at older age, we determined whether the protective actions of exercise are maintained at later ages. We found that during peri-menopause, exercise no longer improves resilience to stress and even becomes anxiogenic. Furthermore, the interaction seen in young females between the E2 α receptor (ERα) and the IGF-I receptor (IGF-IR) is lost at middle-age. In addition, E2 no longer induces IGF-I uptake by brain endothelial cells, and consequently, hippocampal IGF-I levels do not increase. Treatment of middle-aged females with an ERα agonist did not recover the positive actions of exercise. Collectively, these data indicate that the loss of action of exercise during peri-menopause may be related to a loss of the interaction of IGF-IR with ERα in brain endothelial cells that cannot be ameliorated by estrogen therapy. Changes in regulation of mood by physical activity may contribute to increased appearance of affective disorders along age. VM was a recipient of an FPI fellowship and from Ciberned.     Alzheimer’s disease (AD) is an age-dependent neurodegenerative disease constituting ~95% of late-onset non-familial/sporadic AD, and only ~5% accounting for early-onset familial AD. Availability of a pertinent model representing sporadic AD is essential for testing candidate therapies. Emerging evidence indicates a causal link between diabetes and AD. People with diabetes are >1.5-fold more likely to develop AD. Senescence-accelerated mouse model (SAMP8) of accelerated aging displays many features occurring early in AD. Given the role played by diabetes in the pre-disposition of AD, and the utility of SAMP8 non-transgenic mouse model of accelerated aging, we examined if high fat diet-induced experimental type 2 diabetes in SAMP8 mice will trigger pathological aging of the brain. Results showed that compared to non-diabetic SAMP8 mice, diabetic SAMP8 mice exhibited increased cerebral amyloid-β, dysregulated tau-phosphorylating glycogen synthase kinase 3β, reduced synaptophysin immunoreactivity, and displayed memory deficits, indicating Alzheimer-like changes. High fat diet-induced type 2 diabetic SAMP8 mice may represent the metabolic model of AD.

    Abstract. This paper describes the ionospheric response to a geomagnetic storm beginning on 17 April 2002. We present the measurements of ionospheric parameters in the F-region obtained by the network of eight incoherent scatter radars. The main effects of this storm include a deep decrease in the electron density observed at high and middle latitudes in the pre-noon sector, and a minor enhancement in the density observed in the daytime sector at middle latitudes. Extreme plasma heating (>1000-3000 K) is observed at high latitudes, subsiding to 200-300K at subauroral latitudes. The western hemisphere radar chain observed the prompt penetration of the electric field from auroral to equatorial latitudes, as well as the daytime enhancement of plasma drift parallel to the magnetic field line, which is related to the enhancement in the equatorward winds. We suggest that in the first several hours after the storm onset, a negative phase above Millstone Hill (pre-noon sector) results from counteracting processes - penetration electric field, meridional wind, and electrodynamic heating, with electrodynamic heating being the dominant mechanism. At the lower latitude in the pre-noon sector (Arecibo and Jicamarca), the penetration electric field becomes more important, leading to a negative storm phase over Arecibo. In contrast, in the afternoon sector at mid-latitudes (Kharkov, Irkutsk), effects of penetration electric field and meridional wind do not counteract, but add up, leading to a small (~15%), positive storm phase over these locations. As the storm develops, Millstone Hill and Irkutsk mid-latitude radars observe further depletion of electron density due to the changes in the neutral composition.

    Abstract. Taenia solium cysticercosis is difficult to eliminate without interventions or societal development. Atahualpa is a rural Ecuadorian village with documented low migration rate, where domestic pig raising is common and human cysticercosis is endemic. To assess neurocysticercosis (NCC) prevalence, 1,273 villagers aged ≥ 20 years underwent neuroimaging studies, which showed calcified lesions in 121 (9.5%) individuals, but no active disease. Likewise, positive reactions, apparently nonspecific, were found in only 3/200 subjects by the use of a monoclonal antibody-based enzyme-linked immunosorbent assay to detect T. solium antigens in urine. Only 2/418 pigs reacted to three antibody bands on serum western blot and none to more than three bands. This is the first time that spontaneously arrested T. solium transmission is documented in a known endemic village. Understanding why active transmission stopped could provide insights on potential targets for control interventions. Atahualpa could provide an optimal scenario for longitudinal studies on the consequences of calcified NCC.

    Abstract Conditioned pain modulation (CPM) can discriminate between healthy and chronic pain patients. However, its relationship with neurophysiological pain mechanisms is poorly understood. Brain oscillations measured by electroencephalography (EEG) might help gain insight into this complex relationship.ObjectiveTo investigate the relationship between CPM response and self-reported pain intensity in non-specific chronic low back pain (NSCLBP) and explore respective EEG signatures associated to these mechanisms.DesignCross-sectional analysis.ParticipantsThirty NSCLBP patients participated.MethodsSelf-reported low back pain, questionnaires, mood scales, CPM (static and dynamic quantitative sensory tests), and resting surface EEG data were collected and analyzed. Linear regression models were used for statistical analysis.ResultsCPM was not significantly correlated with self-reported pain intensity scores. Relative power of EEG in the beta and high beta bands as recorded from the frontal, central, and parietal cortical areas were significantly associated with CPM. EEG relative power at delta and theta bands as recorded from the central area were significantly correlated with self-reported pain intensity scores while controlling for self-reported depression.ConclusionsFaster EEG frequencies recorded from pain perception areas may provide a signature of a potential cortical compensation caused by chronic pain states. Slower EEG frequencies may have a critical role in abnormal pain processing.

    Electrosyneresis and double diffusion are immunoprecipitation techniques commonly used in the serological diagnosis of Farmer's lung disease (FLD). These techniques are reliable but lack standardization. The aim of this study was to evaluate Western blotting for the serodiagnosis of FLD. We carried out Western blotting with an antigenic extract of Lichtheimia corymbifera, an important aetiological agent of the disease. The membranes were probed with sera from 21 patients with FLD and 21 healthy exposed controls to examine the IgG antibody responses against purified somatic antigens. Given the low prevalence of the disease, 21 patients could be considered as a relevant series. Four bands were significantly more frequently represented in membranes probed with FLD sera (bands at 27.7, 40.5, 44.0 and 50.5 kDa) than those probed with control sera. We assessed the diagnostic value of different criteria alone or in combination. The diagnostic accuracy of the test was highest with the inclusion of at least two of the following criteria: at least five bands on the strip and the presence of one band at 40.5 or 44.0 kDa. Sensitivity, specificity and positive and negative predictive values were all 81%, and the odds ratio was 18.06. Inclusion of bands of high intensity diminished rather than improved the diagnostic value of the test. We concluded that Western blotting is a valuable technique for the serodiagnosis of FLD. The industrial production of ready-to-use membranes would enable the routine use of this technique in laboratories, and provide reliable and standardized diagnostic results within a few hours.

    One of the most severe types of cancer caused by the uncontrollable proliferation of brain cells inside the skull is brain tumors. Hence, a fast and accurate tumor detection method is critical for the patient’s health. Many automated artificial intelligence (AI) methods have recently been developed to diagnose tumors. These approaches, however, result in poor performance; hence, there is a need for an efficient technique to perform precise diagnoses. This paper suggests a novel approach for brain tumor detection via an ensemble of deep and hand-crafted feature vectors (FV). The novel FV is an ensemble of hand-crafted features based on the GLCM (gray level co-occurrence matrix) and in-depth features based on VGG16. The novel FV contains robust features compared to independent vectors, which improve the suggested method’s discriminating capabilities. The proposed FV is then classified using SVM or support vector machines and the k-nearest neighbor classifier (KNN). The framework achieved the highest accuracy of 99% on the ensemble FV. The results indicate the reliability and efficacy of the proposed methodology; hence, radiologists can use it to detect brain tumors through MRI (magnetic resonance imaging). The results show the robustness of the proposed method and can be deployed in the real environment to detect brain tumors from MRI images accurately. In addition, the performance of our model was validated via cross-tabulated data.

    AbstractGuilt, a self-conscious emotion, includes self-focused role taking and also correlates with other-oriented role-taking. Excess guilt proneness might be relevant to obsessive compulsive disorders. The white matter (WM) neural correlates of the degree of guilt have not yet been determined. We hypothesized that the WM structures involved in feelings of guilt are associated with social and moral cognition (inferior parietal lobule [IPL], prefrontal cortex [PFC], and cingulate), and aimed to visualize this using diffusion MRI. We investigated the association between regional WM structures (WM volume, and fractional anisotropy, and mean diffusivity [MD]), and feelings of guilt in 1196 healthy, young students using MRI and the Guilty Feeling Scale, which comprises interpersonal situation (IPS; guilt from hurting friends) and rule-breaking situation (RBS; deontological guilt) scores. The primary novel finding presented here is that MD in the right somatosensory and motor cortices from arm to hand were positively correlated with RBS scores. Further, consistent with our hypothesis, RBS scores were positively correlated with MD in the same regions. These results would be predicted by the Macbeth effect, an obsession with dirt leading to hand-washing rituals resulting from guilt, made famous by the Shakespearian character Lady Macbeth. “What, will these hands ne’er be clean?” William Shakespeare (Shakespeare, 1606) Macbeth.

    Although exact causes of Parkinson disease (PD) remain enigmatic, mitochondrial dysfunction is increasingly appreciated as a key determinant of dopaminergic neuron susceptibility in both familial and sporadic PD. Two genes associated with recessive, early-onset PD encode the ubiquitin (Ub) kinase PINK1 and the E3 Ub ligase PRKN/PARK2/Parkin, which together orchestrate a protective mitochondrial quality control (mitoQC) pathway. Upon stress, both enzymes cooperatively identify and decorate damaged mitochondria with phosphorylated poly-Ub (p-S65-Ub) chains. This specific label is subsequently recognized by autophagy receptors that further facilitate mitochondrial degradation in lysosomes (mitophagy). Here, we analyzed human post-mortem brain specimens and identified distinct pools of p-S65-Ub-positive structures that partially colocalized with markers of mitochondria, autophagy, lysosomes and/or granulovacuolar degeneration bodies. We further quantified levels and distribution of the ‘mitophagy tag’ in 2 large cohorts of brain samples from normal aging and Lewy body disease (LBD) cases using unbiased digital pathology. Somatic p-S65-Ub structures independently increased with age and disease in distinct brain regions and enhanced levels in LBD brain were age- and Braak tangle stage-dependent. Additionally, we observed significant correlations of p-S65-Ub with LBs and neurofibrillary tangle levels in disease. The degree of co-existing p-S65-Ub signals and pathological PD hallmarks increased in the pre-mature stage, but decreased in the late stage of LB or tangle aggregation. Altogether, our study provides further evidence for a potential pathogenic overlap among different forms of PD and suggests that p-S65-Ub can serve as a biomarker for mitochondrial damage in aging and disease. 11,059 4,168 1418 1404 SCIE Q1

    Antiparasitic agents against Taenia solium cysticercosis have been in use since 1979, although its use has been questioned on the basis that cysts would die naturally and thus treatment-induced inflammation is unnecessary. In addition, isolated reports have also questioned whether neurocysticercosis (NCC) is a cause of epilepsy. After more than three and a half decades, a large body of evidence is available. Little if any doubt exists about NCC as a cause of seizures - NCC is consistently associated with seizures when appropriate groups are compared, and in a large subset of cases, seizure semiology correlates with the anatomical location of lesions. Cyst degeneration and the subsequent inflammatory reaction increase seizure expression, although patients with non-inflamed cysts may have seizures, as do patients with long-standing, not inflamed calcified scars. Assessment of the evidence on cysticidal efficacy, safety, and the impact of cyst destruction in decreasing seizures leads to the conclusion that the benefits of antiparasitic treatment in parenchymal brain cysticercosis clearly outweigh the risks, and have provided substantive evidence of the role of NCC as a cause of seizures and epilepsy. Antiparasitic therapy should be considered a primary option in the management of patients with live or degenerating brain NCC cysts.     We recently reported that exercise increases resilience to stress in young female mice. Underlying mechanisms include an interaction of the ovarian hormone estradiol (E2) with insulin-like growth factor I (IGF-I), and an increase in the hippocampal levels of the latter. Since changes in mood regulation during aging may contribute to increasing incidence of affective disorders at older age, we determined whether the protective actions of exercise are maintained at later ages. We found that during peri-menopause, exercise no longer improves resilience to stress and even becomes anxiogenic. Furthermore, the interaction seen in young females between the E2 α receptor (ERα) and the IGF-I receptor (IGF-IR) is lost at middle-age. In addition, E2 no longer induces IGF-I uptake by brain endothelial cells, and consequently, hippocampal IGF-I levels do not increase. Treatment of middle-aged females with an ERα agonist did not recover the positive actions of exercise. Collectively, these data indicate that the loss of action of exercise during peri-menopause may be related to a loss of the interaction of IGF-IR with ERα in brain endothelial cells that cannot be ameliorated by estrogen therapy. Changes in regulation of mood by physical activity may contribute to increased appearance of affective disorders along age. VM was a recipient of an FPI fellowship and from Ciberned.     Alzheimer’s disease (AD) is an age-dependent neurodegenerative disease constituting ~95% of late-onset non-familial/sporadic AD, and only ~5% accounting for early-onset familial AD. Availability of a pertinent model representing sporadic AD is essential for testing candidate therapies. Emerging evidence indicates a causal link between diabetes and AD. People with diabetes are >1.5-fold more likely to develop AD. Senescence-accelerated mouse model (SAMP8) of accelerated aging displays many features occurring early in AD. Given the role played by diabetes in the pre-disposition of AD, and the utility of SAMP8 non-transgenic mouse model of accelerated aging, we examined if high fat diet-induced experimental type 2 diabetes in SAMP8 mice will trigger pathological aging of the brain. Results showed that compared to non-diabetic SAMP8 mice, diabetic SAMP8 mice exhibited increased cerebral amyloid-β, dysregulated tau-phosphorylating glycogen synthase kinase 3β, reduced synaptophysin immunoreactivity, and displayed memory deficits, indicating Alzheimer-like changes. High fat diet-induced type 2 diabetic SAMP8 mice may represent the metabolic model of AD.

    Abstract. This paper describes the ionospheric response to a geomagnetic storm beginning on 17 April 2002. We present the measurements of ionospheric parameters in the F-region obtained by the network of eight incoherent scatter radars. The main effects of this storm include a deep decrease in the electron density observed at high and middle latitudes in the pre-noon sector, and a minor enhancement in the density observed in the daytime sector at middle latitudes. Extreme plasma heating (>1000-3000 K) is observed at high latitudes, subsiding to 200-300K at subauroral latitudes. The western hemisphere radar chain observed the prompt penetration of the electric field from auroral to equatorial latitudes, as well as the daytime enhancement of plasma drift parallel to the magnetic field line, which is related to the enhancement in the equatorward winds. We suggest that in the first several hours after the storm onset, a negative phase above Millstone Hill (pre-noon sector) results from counteracting processes - penetration electric field, meridional wind, and electrodynamic heating, with electrodynamic heating being the dominant mechanism. At the lower latitude in the pre-noon sector (Arecibo and Jicamarca), the penetration electric field becomes more important, leading to a negative storm phase over Arecibo. In contrast, in the afternoon sector at mid-latitudes (Kharkov, Irkutsk), effects of penetration electric field and meridional wind do not counteract, but add up, leading to a small (~15%), positive storm phase over these locations. As the storm develops, Millstone Hill and Irkutsk mid-latitude radars observe further depletion of electron density due to the changes in the neutral composition.

    Abstract. Taenia solium cysticercosis is difficult to eliminate without interventions or societal development. Atahualpa is a rural Ecuadorian village with documented low migration rate, where domestic pig raising is common and human cysticercosis is endemic. To assess neurocysticercosis (NCC) prevalence, 1,273 villagers aged ≥ 20 years underwent neuroimaging studies, which showed calcified lesions in 121 (9.5%) individuals, but no active disease. Likewise, positive reactions, apparently nonspecific, were found in only 3/200 subjects by the use of a monoclonal antibody-based enzyme-linked immunosorbent assay to detect T. solium antigens in urine. Only 2/418 pigs reacted to three antibody bands on serum western blot and none to more than three bands. This is the first time that spontaneously arrested T. solium transmission is documented in a known endemic village. Understanding why active transmission stopped could provide insights on potential targets for control interventions. Atahualpa could provide an optimal scenario for longitudinal studies on the consequences of calcified NCC.

    Abstract Conditioned pain modulation (CPM) can discriminate between healthy and chronic pain patients. However, its relationship with neurophysiological pain mechanisms is poorly understood. Brain oscillations measured by electroencephalography (EEG) might help gain insight into this complex relationship.ObjectiveTo investigate the relationship between CPM response and self-reported pain intensity in non-specific chronic low back pain (NSCLBP) and explore respective EEG signatures associated to these mechanisms.DesignCross-sectional analysis.ParticipantsThirty NSCLBP patients participated.MethodsSelf-reported low back pain, questionnaires, mood scales, CPM (static and dynamic quantitative sensory tests), and resting surface EEG data were collected and analyzed. Linear regression models were used for statistical analysis.ResultsCPM was not significantly correlated with self-reported pain intensity scores. Relative power of EEG in the beta and high beta bands as recorded from the frontal, central, and parietal cortical areas were significantly associated with CPM. EEG relative power at delta and theta bands as recorded from the central area were significantly correlated with self-reported pain intensity scores while controlling for self-reported depression.ConclusionsFaster EEG frequencies recorded from pain perception areas may provide a signature of a potential cortical compensation caused by chronic pain states. Slower EEG frequencies may have a critical role in abnormal pain processing.

    Electrosyneresis and double diffusion are immunoprecipitation techniques commonly used in the serological diagnosis of Farmer's lung disease (FLD). These techniques are reliable but lack standardization. The aim of this study was to evaluate Western blotting for the serodiagnosis of FLD. We carried out Western blotting with an antigenic extract of Lichtheimia corymbifera, an important aetiological agent of the disease. The membranes were probed with sera from 21 patients with FLD and 21 healthy exposed controls to examine the IgG antibody responses against purified somatic antigens. Given the low prevalence of the disease, 21 patients could be considered as a relevant series. Four bands were significantly more frequently represented in membranes probed with FLD sera (bands at 27.7, 40.5, 44.0 and 50.5 kDa) than those probed with control sera. We assessed the diagnostic value of different criteria alone or in combination. The diagnostic accuracy of the test was highest with the inclusion of at least two of the following criteria: at least five bands on the strip and the presence of one band at 40.5 or 44.0 kDa. Sensitivity, specificity and positive and negative predictive values were all 81%, and the odds ratio was 18.06. Inclusion of bands of high intensity diminished rather than improved the diagnostic value of the test. We concluded that Western blotting is a valuable technique for the serodiagnosis of FLD. The industrial production of ready-to-use membranes would enable the routine use of this technique in laboratories, and provide reliable and standardized diagnostic results within a few hours.
