Ferguson;Very low‐frequency blood oxygen level‐dependent (BOLD) fluctuations have emerged as a valuable tool for describing brain anatomy, neuropathology, and development. Such fluctuations exhibit power law frequency dynamics, with largest amplitude at lowest frequencies. The biophysical mechanisms generating such fluctuations are poorly understood. Using publicly available data from 1,019 subjects of age 7–30, we show that BOLD fluctuations exhibit temporal complexity that is linearly related to local connectivity (regional homogeneity), consistently and significantly covarying across subjects and across gray matter regions. This relationship persisted independently of covariance with gray matter density or standard deviation of BOLD signal. During late neurodevelopment, BOLD fluctuations were unchanged with age in association cortex while becoming more random throughout the rest of the brain. These data suggest that local interconnectivity may play a key role in establishing the complexity of low‐frequency BOLD fluctuations underlying functional magnetic resonance imaging connectivity. Stable low‐frequency power dynamics may emerge through segmentation and integration of connectivity during development of distributed large‐scale brain networks. We examined whether violation of non-local dependencies manifest in event related potentials (ERP) for Western and Iranian excerpts by recording EEG while participants passively listened to sequences of modified/original excerpts. We also investigated oscillatory and synchronization properties of brain responses during processing of hierarchical structures. For the Western excerpt, subjective ratings of conclusiveness were marginally significant and the difference in the ERP components fell short of significance. However, ERP and behavioral results showed that while listening to culturally familiar music, subjects comprehended whether or not the hierarchical syntactic structure was fulfilled. Irregularities in the hierarchical structures of the Iranian excerpt elicited an early negativity in the central regions bilaterally, followed by two later negativities from 450–700 to 750–950 ms. The latter manifested throughout the scalp. Moreover, violations of hierarchical structure in the Iranian excerpt were associated with (i) an early decrease in the long range alpha phase synchronization, (ii) an early increase in the oscillatory activity in the beta band over the central areas, and (iii) a late decrease in the theta band phase synchrony between left anterior and right posterior regions. Results suggest that rhythmic structures and melodic fragments, representative of Iranian music, created a familiar context in which recognition of complex non-local syntactic structures was feasible for Iranian listeners. Processing of neural responses to the Iranian excerpt indicated neural mechanisms for processing of hierarchical syntactic structures in music at different levels of cortical integration.

Langeslag;A preference for organization is associated with several disorders, but is widespread in the general population as well. It remains unclear whether organization and various degrees of disorganization elicit pleasant or unpleasant feelings (i.e., valence), calming or arousing feelings (i.e., arousal), and a frontal negativity in the event-related potential (ERP) related to cognitive control. This study tested how organization, slight disorganization, and total disorganization affect valence, arousal, and the frontal negativity. Participants passively viewed organized, slightly disorganized, totally disorganized, and control pictures while their electroencephalogram was recorded. They also rated the valence and arousal elicited by each picture and completed questionnaires assessing desire for order and organization behavior. Organized pictures made participants feel most pleasant, control pictures made participants feel less pleasant, slightly disorganized pictures made participants feel even less pleasant, and totally disorganized pictures made participants feel least pleasant. There were no significant effects on arousal. Totally disorganized pictures elicited a frontal negativity in the ERP between 200-2000 ms after stimulus onset, which might reflect inhibition of rearranging behavior. Individual differences in desire for order and organization behavior did not correlate with valence, arousal, or the frontal negativity. The current study design and findings could be a starting point for examining the differences between adaptive and maladaptive preferences for organization and aversions to disorganization.

Styner; Martin A. Styner;The infant brain undergoes a remarkable period of neural development that is crucial for the development of cognitive and behavioral capacities (Hasegawa et al., 2018). Longitudinal magnetic resonance imaging (MRI) is able to characterize the developmental trajectories and is critical in neuroimaging studies of early brain development. However, missing data at different time points is an unavoidable occurrence in longitudinal studies owing to participant attrition and scan failure. Compared to dropping incomplete data, data imputation is considered a better solution to address such missing data in order to preserve all available samples. In this paper, we adapt generative adversarial networks (GAN) to a new application: longitudinal image prediction of structural MRI in the first year of life. In contrast to existing medical image-to-image translation applications of GANs, where inputs and outputs share a very close anatomical structure, our task is more challenging as brain size, shape and tissue contrast vary significantly between the input data and the predicted data. Several improvements over existing GAN approaches are proposed to address these challenges in our task. To enhance the realism, crispness, and accuracy of the predicted images, we incorporate both a traditional voxel-wise reconstruction loss as well as a perceptual loss term into the adversarial learning scheme. As the differing contrast changes in T1w and T2w MR images in the first year of life, we incorporate multi-contrast images leading to our proposed 3D multi-contrast perceptual adversarial network (MPGAN). Extensive evaluations are performed to assess the qualityand fidelity of the predicted images, including qualitative and quantitative assessments of the image appearance, as well as quantitative assessment on two segmentation tasks. Our experimental results show that our MPGAN is an effective solution for longitudinal MR image data imputation in the infant brain. We further apply our predicted/imputed images to two practical tasks, a regression task and a classification task, in order to highlight the enhanced task-related performance following image imputation. The results show that the model performance in both tasks is improved by including the additional imputed data, demonstrating the usability of the predicted images generated from our approach.

McIntyre;<i>Objective:</i> Deep brain stimulation (DBS) surgeries commonly rely on brain atlases and microelectrode recordings (MER) to help identify the target location for electrode implantation. We present an automated method for optimally fitting a 3-dimensional brain atlas to intraoperative MER and predicting a target DBS electrode location in stereotactic coordinates for the patient. <i>Methods:</i> We retrospectively fit a 3-dimensional brain atlas to MER points from 10 DBS surgeries targeting the subthalamic nucleus (STN). We used a constrained optimization algorithm to maximize the MER points correctly fitted (i.e., contained) within the appropriate atlas nuclei. We compared our optimization approach to conventional anterior commissure-posterior commissure (AC/PC) scaling, and to manual fits performed by four experts. A theoretical DBS electrode target location in the dorsal STN was customized to each patient as part of the fitting process and compared to the location of the clinically defined therapeutic stimulation contact. <i>Results:</i> The human expert and computer optimization fits achieved significantly better fits than the AC/PC scaling (80, 81, and 41% of correctly fitted MER, respectively). However, the optimization fits were performed in less time than the expert fits and converged to a single solution for each patient, eliminating interexpert variance. <i>Conclusions and Significance:</i> DBS therapeutic outcomes are directly related to electrode implantation accuracy. Our automated fitting techniques may aid in the surgical decision-making process by optimally integrating brain atlas and intraoperative neurophysiological data to provide a visual guide for target identification.

Salzer; WP Holmes; Colman;The myelin associated glycoproteins (MAG) are integral plasma membrane proteins which are found in oligodendrocytes and Schwann cells and are believed to mediate the axonal-glial interactions of myelination. In this paper we demonstrate the existence in central nervous system myelin of two MAG polypeptides with Mrs of 67,000 and 72,000 that we have designated small MAG (S-MAG) and large MAG (L-MAG), respectively. The complete amino acid sequence of L-MAG and a partial amino acid sequence of S-MAG have been deduced from the nucleotide sequences of corresponding cDNA clones isolated from a lambda gt11 rat brain expression library. Based on their amino acid sequences, we predict that both proteins have an identical membrane spanning segment and a large extracellular domain. The putative extracellular region contains an Arg-Gly-Asp sequence that may be involved in the interaction of these proteins with the axon. The extracellular portion of L-MAG also contains five segments of internal homology that resemble immunoglobulin domains, and are strikingly homologous to similar domains of the neural cell adhesion molecule and other members of the immunoglobulin gene superfamily. In addition, the two MAG proteins differ in the extent of their cytoplasmically disposed segments and appear to be the products of alternatively spliced mRNAs. Of considerable interest is the finding that the cytoplasmic domain of L-MAG, but not of S-MAG, contains an amino acid sequence that resembles the autophosphorylation site of the epidermal growth factor receptor.

These sources are representative of the brightest ~10% of the Cosmic Infrared Background. They are a mixture of mostly local (z<~0.3) starforming galaxies, and a tail of ULIGs that extend up to z~1, and are likely to be a similar population to faint SCUBA sources. We use archival Spitzer IRAC and MIPS data to extend the spectral coverage to the mid-IR regime, resulting in an unprecended (for this redshift range) census of their infrared SEDs. This allows us to study in far greater detail this important population linking the near-IR stellar emission with PAH and thermal dust emission. We do this using a Markov Chain Monte Carlo method, which easily allows for the inclusion of ~6 free parameters, as well as an estimate of parameter uncertainties and correlations. Miller;Zhihua Xie; Gregory M. Miller;This study reveals a novel receptor mechanism for methamphetamine action in dopamine transporter (DAT) regulation. Trace amine-associated receptor 1 (TAAR1) is expressed in brain dopaminergic nuclei and is activated by methamphetamine in vitro. Here, we show that methamphetamine interaction with TAAR1 inhibits [3H]dopamine uptake, enhances or induces [3H]dopamine efflux, and triggers DAT internalization. In time course assays in which methamphetamine and [3H]dopamine were concurrently loaded into cells or synaptosomes or in pretreatment assays in which methamphetamine was washed away before [3H]dopamine loading, methamphetamine caused a distinct inhibition in [3H]dopamine uptake in TAAR1 + DAT-cotransfected cells and in wild-type mouse and rhesus monkey striatal synaptosomes. This distinct uptake inhibition was not observed in DAT-only transfected cells or in TAAR1 knockout mouse striatal synaptosomes. In [3H]dopamine efflux assays using the same cell and synaptosome preparations, methamphetamine enhanced [3H]dopamine efflux at a high loading concentration of [3H]dopamine (1 μM) or induced [3H]dopamine efflux at a low loading concentration of [3H]dopamine (10 nM) in a TAAR1-dependent manner. In DAT biotinylation assays using the same cell and synaptosome preparations, we observed that 1 μM methamphetamine induced DAT internalization in a TAAR1-dependent manner. All these TAAR1-mediated effects of methamphetamine were blocked by the protein kinase inhibitors H89 [N-[2-(4-bromocinnamylamino)ethyl]-5-isoquinoline] and/or 2-{8-[(dimethylamino) methyl]-6,7,8,9-tetrahydropyrido[1,2-a]indol-3-yl}-3-(1-methylindol-3-yl)maleimide (Ro32-0432), suggesting that methamphetamine interaction with TAAR1 triggers cellular phosphorylation cascades and leads to the observed effects of methamphetamine on DAT. These findings demonstrate a mediatory role of TAAR1 in methamphetamine action in DAT regulation and implicate this receptor as a potential target of therapeutics drugs for methamphetamine addiction.

Ortinski; Stefano Vicini;GABA(A) receptor subtypes comprising the alpha1 and alpha3 subunits change with development and have a specific anatomical localization in the adult brain. These receptor subtypes have been previously demonstrated to greatly differ in deactivation kinetics but the underlying gating mechanisms have not been fully elucidated. Therefore, we expressed rat alpha1beta2gamma2 and alpha3beta2gamma2 receptors in human embryonic kidney 293 cells and recorded current responses to ultrafast GABA applications at macroscopic and single-channel levels. We found that the slow deactivation of alpha3beta2gamma2-mediated currents is associated with a relatively small rate and extent of apparent desensitization. In contrast, responses mediated by alpha1beta2gamma2 receptors had faster deactivation and stronger desensitization. Alpha3beta2gamma2 receptors had faster recovery in the paired-pulse agonist applications than alpha1beta2gamma2 channels. The onset of currents mediated by alpha3beta2gamma2 receptors was slower than that of alpha1beta2gamma2 for a wide range of GABA concentrations. Single-channel analysis did not reveal differences in the opening/closing kinetics of alpha1beta2gamma2 and alpha3beta2gamma2 channels but burst durations were longer in alpha3beta2gamma2 receptors. Simulation with a previously reported kinetic model was used to explore the differences in respective rate constants. Reproduction of major kinetic differences required a smaller desensitization rate as well as smaller binding and unbinding rates in alpha3beta2gamma2 compared with alpha1beta2gamma2 receptors. Our work describes the mechanisms underlying the kinetic differences between two major GABA(A) receptor subtypes and provides a framework to interpret data from native GABA receptors.

Emre Koksal; Philip Schniter;In this paper, we address the problem of joint scheduling and resource allocation in the downlink of an orthogonal frequency division multiple access (OFDMA)-based wireless network when the per-user SNR is known only in distribution. In particular, we consider sum-utility maximization over user schedules, powers, and code rates, subject to an instantaneous sum-power constraint. We consider both a “continuous” scenario where, during a time-slot, each OFDMA subchannel can be time-shared among multiple users and/or code rates, and a “discrete” scenario where no time-sharing is allowed. For the non-convex optimization problem arising in the continuous case, we propose an efficient exact solution. For the mixed-integer optimization problem arising in the discrete case, we propose a polynomial-complexity approximate solution and derive a bound on its optimality gap. We also provide a numerical study of goodput maximization for the SNR distribution that results from the use of pilot-aided MMSE channel estimation.

Ferguson;Very low‐frequency blood oxygen level‐dependent (BOLD) fluctuations have emerged as a valuable tool for describing brain anatomy, neuropathology, and development. Such fluctuations exhibit power law frequency dynamics, with largest amplitude at lowest frequencies. The biophysical mechanisms generating such fluctuations are poorly understood. Using publicly available data from 1,019 subjects of age 7–30, we show that BOLD fluctuations exhibit temporal complexity that is linearly related to local connectivity (regional homogeneity), consistently and significantly covarying across subjects and across gray matter regions. This relationship persisted independently of covariance with gray matter density or standard deviation of BOLD signal. During late neurodevelopment, BOLD fluctuations were unchanged with age in association cortex while becoming more random throughout the rest of the brain. These data suggest that local interconnectivity may play a key role in establishing the complexity of low‐frequency BOLD fluctuations underlying functional magnetic resonance imaging connectivity. Stable low‐frequency power dynamics may emerge through segmentation and integration of connectivity during development of distributed large‐scale brain networks. We examined whether violation of non-local dependencies manifest in event related potentials (ERP) for Western and Iranian excerpts by recording EEG while participants passively listened to sequences of modified/original excerpts. We also investigated oscillatory and synchronization properties of brain responses during processing of hierarchical structures. For the Western excerpt, subjective ratings of conclusiveness were marginally significant and the difference in the ERP components fell short of significance. However, ERP and behavioral results showed that while listening to culturally familiar music, subjects comprehended whether or not the hierarchical syntactic structure was fulfilled. Irregularities in the hierarchical structures of the Iranian excerpt elicited an early negativity in the central regions bilaterally, followed by two later negativities from 450–700 to 750–950 ms. The latter manifested throughout the scalp. Moreover, violations of hierarchical structure in the Iranian excerpt were associated with (i) an early decrease in the long range alpha phase synchronization, (ii) an early increase in the oscillatory activity in the beta band over the central areas, and (iii) a late decrease in the theta band phase synchrony between left anterior and right posterior regions. Results suggest that rhythmic structures and melodic fragments, representative of Iranian music, created a familiar context in which recognition of complex non-local syntactic structures was feasible for Iranian listeners. Processing of neural responses to the Iranian excerpt indicated neural mechanisms for processing of hierarchical syntactic structures in music at different levels of cortical integration.

Langeslag;A preference for organization is associated with several disorders, but is widespread in the general population as well. It remains unclear whether organization and various degrees of disorganization elicit pleasant or unpleasant feelings (i.e., valence), calming or arousing feelings (i.e., arousal), and a frontal negativity in the event-related potential (ERP) related to cognitive control. This study tested how organization, slight disorganization, and total disorganization affect valence, arousal, and the frontal negativity. Participants passively viewed organized, slightly disorganized, totally disorganized, and control pictures while their electroencephalogram was recorded. They also rated the valence and arousal elicited by each picture and completed questionnaires assessing desire for order and organization behavior. Organized pictures made participants feel most pleasant, control pictures made participants feel less pleasant, slightly disorganized pictures made participants feel even less pleasant, and totally disorganized pictures made participants feel least pleasant. There were no significant effects on arousal. Totally disorganized pictures elicited a frontal negativity in the ERP between 200-2000 ms after stimulus onset, which might reflect inhibition of rearranging behavior. Individual differences in desire for order and organization behavior did not correlate with valence, arousal, or the frontal negativity. The current study design and findings could be a starting point for examining the differences between adaptive and maladaptive preferences for organization and aversions to disorganization.
