    Herpes simplex virus type 1 (HSV-1) plasmid vectors have promise for genetic intervention in the brain, but several problems caused by the helper virus have compromised their utility. To develop a helper virus-free packaging system for these vectors, the DNA cleavage/packaging signals were deleted from a set of cosmids that represents the HSV-1 genome. Following cotransfection into cells, this modified cosmid set supported replication and packaging of vector DNA. However, in the absence of the DNA cleavage/packaging signals, the HSV-1 genome was not packaged, and consequently vector stocks were free of detectable helper virus. In the absence of helper virus, the vectors efficiently infected rat neural cells in culture or in the brain with minimal cytopathic effects. beta-galactosidase-positive cells were observed for at least 1 month in vivo, and vector DNA persisted for this period. This system may facilitate studies on neuronal physiology and potential therapeutic applications.

    AbstractThe signal loss susceptibility artifact is a major limitation in gradient‐echo MRI applications. Various methods, including z‐shim techniques and multidimensional tailored radio frequency (RF) pulses, have been proposed to mitigate the through‐plane signal loss artifact, which is dominant in axial slices above the sinus region. Unfortunately, z‐shim techniques require multiple steps and multidimensional RF methods are complex, with long pulse lengths. Parallel transmission methods were recently shown to be promising for improving B1 inhomogeneity and reducing the specific absorption rate. In this work, a novel method using time‐shifted slice‐select RF pulses is presented for reducing the through‐plane signal loss artifact in parallel transmission applications. A simultaneous z‐shim is obtained by concurrently applying unique time‐shifted pulses on each transmitter. The method is shown to reduce the signal loss susceptibility artifact in gradient‐echo images using a four‐channel parallel transmission system at 3T.     <b><i>Background:</i></b> Deep brain stimulation (DBS) of the globus pallidus internus is established as efficacious for dystonia, yet the optimal target within this structure is not well defined. Published evidence suggests that spatial normalization provides a better estimate of DBS lead location than traditional methods based on standard stereotactic coordinates. <b><i>Methods:</i></b> We retrospectively reviewed our pallidal implanted dystonia population. Patient imaging scans were morphed into an MRI atlas using a nonlinear image registration algorithm. Active contact locations were projected onto the atlas and clusters analyzed for the degree of variance in two groups: (1) good and poor responders and (2) cervical (CD) and generalized dystonia (GD). <b><i>Results:</i></b> The average active contact location between CD and GD good responders was distinct but not significantly different. The mean active contact for CD poor responders was significantly different from CD responders and GD poor responders in the dorsoventral direction. <b><i>Conclusions:</i></b> A normalized imaging space is arguably more accurate in visualizing postoperative leads. Despite some separation between groups, this data suggests there was not an optimal pallidal target for common dystonia patients. Degrees of variance overlapped due to a large degree of individual target variation. Patient selection may ultimately be the key to maximizing patient outcomes. © 2014 S. Karger AG, Basel

    Mood and anxiety disorders are complex heterogeneous syndromes that manifest in dysfunctions across multiple brain regions, cell types, and circuits. Biomarkers using brain-wide activity patterns in humans have proven useful in distinguishing between disorder subtypes and identifying effective treatments. In order to improve biomarker identification, it is crucial to understand the basic circuitry underpinning brain-wide activity patterns. Leveraging a large repertoire of techniques, animal studies have examined roles of specific cell types and circuits in driving maladaptive behavior. Recent advances in multiregion recording techniques, data-driven analysis approaches, and machine-learning-based behavioral analysis tools can further push the boundary of animal studies and bridge the gap with human studies, to assess how brain-wide activity patterns encode and drive emotional behavior. Together, these efforts will allow identifying more precise biomarkers to enhance diagnosis and treatment.

    Auditory evoked potentials were recorded from 47 subjects ranging in age from 6 to 76 years in order to assess the effects of maturation and aging on the evoked (N1 and P2) and event-related (N2 and P3) components. Because of clear differences in the effects of age on the event-related components between children (less than 15 years of age) and adults the subjects were divided into two populations for analysis. For adults there was a systematic increase in the latency and decrease in amplitude of each component with age. Also the rate of the age-related increase in latency was proportional to the latency of the component. The scalp distributions of both the stimulus-evoked and event-related components were found to vary with age yielding a more nearly equipotential distribution for older subjects. For children the latencies of the event-related components decreased with age. The stimulus-evoked components had latencies which were not significantly different from those predicted from the adult data. In contrast to the adult data, age affected the scalp distributions of the stimulus-evoked components differently than the event-related components. These results suggest an aging process is reflected in the auditory evoked potential which is not the simple inverse of maturational processes. © 1978.

    Neuropsychiatry represents a field of medicine situated at the crossroads of neurology and psychiatry, and deals with the interface of behavioral phenomena driven by brain dysfunction. Psychiatric symptoms are highly prevalent in these conditions, are a major source of disability and diminished quality of life, and potentially represent the target of treatment interventions that stand to significantly decrease the suffering they generate. In this article, the disease paradigm is explained, with particular attention to its role as an organizing principle for the field. Specific diseases including traumatic brain injury, stroke, Parkinson's disease, Alzheimer's disease, multiple sclerosis, and epilepsy are explored in relation to the presentation of multiple psychiatric phenotypes in each, associations with underlying brain pathology, and existing treatment approaches. Finally the article explores the inherent complexities in this area of research and proposes a framework for future work based on the understanding of phenomenology and associated risk factors, the involvement of the rapidly advancing field of neuroscience, and targeted treatment development to serve as a road map for advancement in the field..

    Axotomy of giant lamprey (Petromyzon marinus) central neurons (anterior bulbar cells) close to their somata results in ectopic axon-like sprouting from the dendritic tips. Such sprouts first appear as swellings at the tips of a small subset of dendrites 2-3 weeks after "close" axotomy. We report here that immunocytochemical examination of these swellings reveals a structure and composition that differs from that of conventional growth cones; incipient sprouts contain many highly phosphorylated neurofilaments (NFs), little tubulin, and virtually no stable (acetylated) microtubules (MTs). The dendrites of anterior bulbar cells after close axotomy also show pronounced changes in NF protein and tubulin staining patterns prior to the emergence of sprouts from the dendrites. The amount of tyrosinated tubulin increases greatly; this rise is tightly coupled to the appearance of highly phosphorylated NFs and the loss of nonphosphorylated NFs in the dendrites. Acetylated tubulin is generally reduced after close axotomy and is selectively lost from dendrites that gave rise to sprouts. These changes indicate that an invasion of the dendrites by phosphorylated NFs may be linked to the destabilization of dendritic MTs, and in some dendrites this may lead to a marked loss of stable MTs, which is correlated with the emergence of NF-filled sprouts from the dendritic tips.

    Although tau pathology, behavioral deficits, and neuronal loss are observed in patients with tauopathies, the relationship between these endpoints has not been clearly established. Here we found that rTg4510 mice, which overexpress human mutant tau in the forebrain, develop progressive age-dependent increases in locomotor activity (LMA), which correlates with neurofibrillary tangle (NFT) pathology, hyperphosphorylated tau levels, and brain atrophy. To further clarify the relationship between these endpoints, we treated the rTg4510 mice with either doxycycline to reduce mutant tau expression or an O-GlcNAcase inhibitor Thiamet G, which has been shown to ameliorate tau pathology in animal models. We found that both doxycycline and Thiamet G treatments starting at 2 months of age prevented the progression of hyperactivity, slowed brain atrophy, and reduced brain hyperphosphorylated tau. In contrast, initiating doxycycline treatment at 4 months reduced neither brain hyperphosphorylated tau nor hyperactivity, further confirming the relationship between these measures. Collectively, our results demonstrate a unique behavioral phenotype in the rTg4510 mouse model of tauopathy that strongly correlates with disease progression, and that early interventions which reduce tau pathology ameliorate the progression of the locomotor dysfunction. These findings suggest that better understanding the relationship between locomotor deficits and tau pathology in the rTg4510 model may improve our understanding of the mechanisms underlying behavioral disturbances in patients with tauopathies.

    Herpes simplex virus type 1 (HSV-1) plasmid vectors have promise for genetic intervention in the brain, but several problems caused by the helper virus have compromised their utility. To develop a helper virus-free packaging system for these vectors, the DNA cleavage/packaging signals were deleted from a set of cosmids that represents the HSV-1 genome. Following cotransfection into cells, this modified cosmid set supported replication and packaging of vector DNA. However, in the absence of the DNA cleavage/packaging signals, the HSV-1 genome was not packaged, and consequently vector stocks were free of detectable helper virus. In the absence of helper virus, the vectors efficiently infected rat neural cells in culture or in the brain with minimal cytopathic effects. beta-galactosidase-positive cells were observed for at least 1 month in vivo, and vector DNA persisted for this period. This system may facilitate studies on neuronal physiology and potential therapeutic applications.

    AbstractThe signal loss susceptibility artifact is a major limitation in gradient‐echo MRI applications. Various methods, including z‐shim techniques and multidimensional tailored radio frequency (RF) pulses, have been proposed to mitigate the through‐plane signal loss artifact, which is dominant in axial slices above the sinus region. Unfortunately, z‐shim techniques require multiple steps and multidimensional RF methods are complex, with long pulse lengths. Parallel transmission methods were recently shown to be promising for improving B1 inhomogeneity and reducing the specific absorption rate. In this work, a novel method using time‐shifted slice‐select RF pulses is presented for reducing the through‐plane signal loss artifact in parallel transmission applications. A simultaneous z‐shim is obtained by concurrently applying unique time‐shifted pulses on each transmitter. The method is shown to reduce the signal loss susceptibility artifact in gradient‐echo images using a four‐channel parallel transmission system at 3T.
