    ABSTRACTBackgroundChildren with neurodevelopmental disorders such as Attention Deficit Hyperactivity Disorder (ADHD) often struggle with behavioral self-regulation (BR), which is associated with daily-life challenges. ADHD sometimes presents with Developmental Coordination Disorder (DCD), but little is known about BR in DCD. BR is thought to involve limbic, prefrontal, parietal and temporal brain areas. Given the risk for negative outcomes, gaining a better understanding of the brain mechanisms underlying BR in children with ADHD and/or DCD is imperative.MethodsResting-state fMRI data collected from 115 children (31 typically developing (TD), 35 ADHD, 21 DCD, 28 ADHD-DCD) aged 7-17 years were preprocessed and motion was mitigated using ICA-AROMA. Emotion control, inhibition, and shifting were assessed as subdomains of BR. Functional connectivity (FC) maps were computed for ten limbic, prefrontal, parietal and temporal regions of interest and were investigated for associations with BR subdomains across all participants as well as for significant group differences.ResultsMultiple FC patterns were associated with BR across all participants. Some FC patterns were associated with multiple BR subdomains, while others were associated with only one. Differences in BR were found only between children with ADHD (i.e. ADHD and ADHD-DCD) and those without ADHD (i.e. TD and DCD). FC differences were also found between children with and without ADHD.ConclusionsOur results show dimensional associations between BR subdomain scores and whole-brain FC and highlight the potential of these associative patterns as brain-based signatures of BR in children with and without ADHD.

    A clinical tool that can diagnose psychiatric illness using functional or structural magnetic resonance (MR) brain images has the potential to greatly assist physicians and improve treatment efficacy. Working toward the goal of automated diagnosis, we propose an approach for automated classification of ADHD and autism based on histogram of oriented gradients (HOG) features extracted from MR brain images, as well as personal characteristic data features. We describe a learning algorithm that can produce effective classifiers for ADHD and autism when run on two large public datasets. The algorithm is able to distinguish ADHD from control with hold-out accuracy of 69.6% (over baseline 55.0%) using personal characteristics and structural brain scan features when trained on the ADHD-200 dataset (769 participants in training set, 171 in test set). It is able to distinguish autism from control with hold-out accuracy of 65.0% (over baseline 51.6%) using functional images with personal characteristic data when trained on the Autism Brain Imaging Data Exchange (ABIDE) dataset (889 participants in training set, 222 in test set). These results outperform all previously presented methods on both datasets. To our knowledge, this is the first demonstration of a single automated learning process that can produce classifiers for distinguishing patients vs. controls from brain imaging data with above-chance accuracy on large datasets for two different psychiatric illnesses (ADHD and autism). Working toward clinical applications requires robustness against real-world conditions, including the substantial variability that often exists among data collected at different institutions. It is therefore important that our algorithm was successful with the large ADHD-200 and ABIDE datasets, which include data from hundreds of participants collected at multiple institutions. While the resulting classifiers are not yet clinically relevant, this work shows that there is a signal in the (f)MRI data that a learning algorithm is able to find. We anticipate this will lead to yet more accurate classifiers, over these and other psychiatric disorders, working toward the goal of a clinical tool for high accuracy differential diagnosis.

    Arithmetic reasoning activates the occipital cortex of congenitally blind people (CB). This activation of visual areas may highlight the functional flexibility of occipital regions deprived of their dominant inputs or relate to the intrinsic computational role of specific occipital regions. We contrasted these competing hypotheses by characterizing the brain activity of CB and sighted participants while performing subtraction, multiplication and a control letter task. In both groups, subtraction selectively activated a bilateral dorsal network commonly activated during spatial processing. Multiplication triggered activity in temporal regions thought to participate in memory retrieval. No between-group difference was observed for the multiplication task whereas subtraction induced enhanced activity in the right dorsal occipital cortex of the blind individuals only. As this area overlaps with regions showing selective tuning to auditory spatial processing and exhibits increased functional connectivity with a dorsal "spatial" network, our results suggest that the recruitment of occipital regions during high-level cognition in the blind actually relates to the intrinsic computational role of the activated regions.

    We used a robotic exoskeleton to quantify specific patterns of abnormal upper limb motor behaviour in people who have had transient ischemic attack (TIA). A cohort of people with TIA was recruited within two weeks of symptom onset. All individuals completed a robotic-based assessment of 8 behavioural tasks related to upper limb motor and proprioceptive function, as well as cognitive function. Robotic task performance was compared to a large cohort of controls without neurological impairments corrected for the influence of age. Impairment in people with TIA was defined as performance below the 5th percentile of controls. Participants with TIA were also assessed with the National Institutes of Health Stroke Scale (NIHSS) score, Chedoke-McMaster Stroke Assessment (CMSA) of the arm, the Behavioural Inattention Test (BIT), the Purdue pegboard test (PPB), and the Montreal Cognitive Assessment (MoCA). Age-related white matter change (ARWMC), prior infarction and cella-media index (CMI) were assessed from baseline CT scan that was performed within 24 hours of TIA. Acute infarction was assessed from diffusion-weighted imaging in a subset of people with TIA. Twenty-two people with TIA were assessed. Robotic assessment showed impaired upper limb motor function in 7/22 people with TIA patients and upper limb sensory impairment in 4/22 individuals. Cognitive tasks involving robotic assessment of the upper limb were completed in 13 participants, of whom 8 (61.5%) showed significant impairment. Abnormal performance in the CMSA arm inventory was present in 12/22 (54.5%) participants. ARWMC was 11.8 ± 6.4 and CMI was 5.4 ± 1.5. DWI was positive in 0 participants. Quantitative robotic assessment showed that people who have had a TIA display a spectrum of upper limb motor and sensory performance deficits as well as cognitive function deficits despite resolution of symptoms and no evidence of tissue infarction.

    Most experimental studies of decision-making have specifically examined situations in which a single correct answer exists (externally guided decision-making). Along with such externally guided decision-making, there are instances of decision-making in which no correct answer based on external circumstances is available for the subject (internally guided decision-making, e.g. preference judgment). We compared these two different types of decision-making in terms of conflict-monitoring and their relation with resting-state brain activity. Current electroencephalography (EEG) data demonstrated that conflict-related N2 amplitudes (i.e., difference between large-conflict and small-conflict conditions) in externally guided decision-making were modulated by the type of external stimulus (i.e., large-conflict stimulus pair or small-conflict stimulus pair) but were not found to be correlated with resting-state brain activity (i.e. resting-state EEG power). In contrast, conflict-related N2 amplitudes in internally guided decision-making were found to be correlated with resting-state brain activity, but were not found to be modulated by the type of stimulus itself: the degree to which the type of external stimulus modulates the conflict during stimulus encoding varies according to individual differences in intrinsic brain activity. Considering those results comprehensively, we demonstrate for the first time resting-state and stimulus-related differences between externally and internally guided decision-making.

    In philosophy, the criteria for personhood (PH) at a specific point in time (synchronic), and the necessary and sufficient conditions of personal identity (PI) over time (diachronic) are traditionally separated. Hence, the transition between both timescales of a person's life remains largely unclear. Personal habits reflect a decision-making (DM) process that binds together synchronic and diachronic timescales. Despite the fact that the actualization of habits takes place synchronically, they presuppose, for the possibility of their generation, time in a diachronic sense. The acquisition of habits therefore rests upon PI over time; that is, the temporal extension of personal decisions is the necessary condition for the possible development of habits. Conceptually, habits can thus be seen as a bridge between synchronic and diachronic timescales of a person's life. In order to investigate the empirical mediation of this temporal linkage, we draw upon the neuronal mechanisms underlying DM; in particular on the distinction between internally and externally guided DM. Externally guided DM relies on external criteria at a specific point in time (synchronic); on a neural level, this has been associated with lateral frontal and parietal brain regions. In contrast, internally guided DM is based on the person's own preferences that involve a more longitudinal and thus diachronic timescale, which has been associated with the brain's intrinsic activity. Habits can be considered to reflect a balance between internally and externally guided DM, which implicates a particular temporal balance between diachronic and synchronic elements, thus linking two different timescales. Based on such evidence, we suggest a habit-based neurophilosophical approach of PH and PI by focusing on the empirically-based linkage between the synchronic and diachronic elements of habits. By doing so, we propose to link together what philosophically has been described and analyzed separately as PH and PI.

    Until recently, neuroimaging data for a research study needed to be collected within one’s own lab. However, when studying inter-individual differences in brain structure, a large sample of participants is necessary. Given the financial costs involved in collecting neuroimaging data from hundreds or thousands of participants, large-scale studies of brain morphology could previously only be conducted by well-funded laboratories with access to MRI facilities and to large samples of participants. With the advent of broad open-access data-sharing initiatives, this has recently changed–here the primary goal of the study is to collect large datasets to be shared, rather than sharing of the data as an afterthought. This paradigm shift is evident as increase in the pace of discovery, leading to a rapid rate of advances in our characterization of brain structure. The utility of open-access brain morphology data is numerous, ranging from observing novel patterns of age-related differences in subcortical structures to the development of more robust cortical parcellation atlases, with these advances being translatable to improved methods for characterizing clinical disorders (see Figure 1 for an illustration). Moreover, structural MRIs are generally more robust than functional MRIs, relative to potential artifacts and in being not task-dependent, resulting in large potential yields. While the benefits of open-access data have been discussed more broadly within the field of cognitive neuroscience elsewhere (Gilmore et al., in press; Poldrack and Gorgolewski, 2014; Van Horn and Gazzaniga, 2013; Van Horn and Toga, 2014; Vogelstein et al., 2016; Voytek, 2016), as well as in other fields (Ascoli et al., 2017; Choudhury et al., 2014; Davies et al., 2017), this opinion paper is focused specifically on the implications of open data to brain morphology research.

    Abstract Background It is currently unclear whether the function of brain regions associated with executive cognitive processing are independently associated with reduced physiological falls risk. If these are related, it would suggest that the development of interventions targeted at improving executive neurocognitive function would be an effective new approach for reducing physiological falls risk in seniors. Methods We performed a secondary analysis of 73 community-dwelling senior women aged 65 to 75 years old who participated in a 12-month randomized controlled trial of resistance training. Functional MRI data were acquired while participants performed a modified Eriksen Flanker Task - a task of selective attention and conflict resolution. Brain volumes were obtained using MRI. Falls risk was assessed using the Physiological Profile Assessment (PPA). Results After accounting for baseline age, experimental group, baseline PPA score, and total baseline white matter brain volume, baseline activation in the left frontal orbital cortex extending towards the insula was negatively associated with reduced physiological falls risk over the 12-month period. In contrast, baseline activation in the paracingulate gyrus extending towards the anterior cingulate gyrus was positively associated with reduced physiological falls risk. Conclusions Baseline activation levels of brain regions underlying response inhibition and selective attention were independently associated with reduced physiological falls risk. This suggests that falls prevention strategies may be facilitated by incorporating intervention components - such as aerobic exercise - that are specifically designed to induce neurocognitive plasticity.     Background: Understanding how practice mediates the transition of brain-behavior networks between early and later stages of learning is constrained by the common approach to analysis of fMRI data. Prior imaging studies have mostly relied on a single scan, and parametric, task-related analyses. Our experiment incorporates a multisession fMRI lexicon-learning experiment with multivariate, whole-brain analysis to further knowledge of the distributed networks supporting practice-related learning in schizophrenia. Methods: Participants with schizophrenia were compared with healthy control participants as they learned a novel lexicon during two fMRI scans over a several-day period. All participants were trained to equal task proficiency prior to scanning. Behavioral-Partial Least Squares, a multivariate analytic approach was used to analyse the imaging data. Permutation testing was used to determine statistical significance; bootstrap resampling was used to determine the reliability of the findings. Results: With practice, healthy control participants transitioned to a brain-accuracy network incorporating dorsostriatal regions in late learning stages. The schizophrenia participants did not transition to this pattern despite comparable behavioral results. Instead, successful learners were differentiated primarily on the basis of greater engagement of perceptual and perceptual-integration brain regions Conclusions: There is a different spatiotemporal unfolding of brain-learning relationships in schizophrenia. In schizophrenia, given the same amount of practice, the movement from networks suggestive of effortful learning towards subcortically-driven procedural ones differs from healthy control participants. Learning performance in schizophrenia being driven instead by varying levels of engagement in perceptual regions suggests that perception itself is impaired in the illness and may impact downstream, ‘higher level’ cognitive impairment.

    Abstract Background In the Canadian Alliance for Healthy Hearts and Minds (CAHHM) cohort, participants underwent magnetic resonance imaging (MRI) of the brain, heart, and abdomen, that generated incidental findings (IFs). The approach to managing these unexpected results remain a complex issue. Our objectives were to describe the CAHHM policy for the management of IFs, to understand the impact of disclosing IFs to healthy research participants, and to reflect on the ethical obligations of researchers in future MRI studies. Methods Between 2013 and 2019, 8252 participants (mean age 58 ± 9 years, 54% women) were recruited with a follow-up questionnaire administered to 909 participants (40% response rate) at 1-year. The CAHHM policy followed a restricted approach, whereby routine feedback on IFs was not provided. Only IFs of severe structural abnormalities were reported. Results Severe structural abnormalities occurred in 8.3% (95% confidence interval 7.7–8.9%) of participants, with the highest proportions found in the brain (4.2%) and abdomen (3.1%). The majority of participants (97%) informed of an IF reported no change in quality of life, with 3% of participants reporting that the knowledge of an IF negatively impacted their quality of life. Furthermore, 50% reported increased stress in learning about an IF, and in 95%, the discovery of an IF did not adversely impact his/her life insurance policy. Most participants (90%) would enrol in the study again and perceived the MRI scan to be beneficial, regardless of whether they were informed of IFs. While the implications of a restricted approach to IF management was perceived to be mostly positive, a degree of diagnostic misconception was present amongst participants, indicating the importance of a more thorough consent process to support participant autonomy. Conclusion The management of IFs from research MRI scans remain a challenging issue, as participants may experience stress and a reduced quality of life when IFs are disclosed. The restricted approach to IF management in CAHHM demonstrated a fair fulfillment of the overarching ethical principles of respect for autonomy, concern for wellbeing, and justice.     ABSTRACTBackgroundChildren with neurodevelopmental disorders such as Attention Deficit Hyperactivity Disorder (ADHD) often struggle with behavioral self-regulation (BR), which is associated with daily-life challenges. ADHD sometimes presents with Developmental Coordination Disorder (DCD), but little is known about BR in DCD. BR is thought to involve limbic, prefrontal, parietal and temporal brain areas. Given the risk for negative outcomes, gaining a better understanding of the brain mechanisms underlying BR in children with ADHD and/or DCD is imperative.MethodsResting-state fMRI data collected from 115 children (31 typically developing (TD), 35 ADHD, 21 DCD, 28 ADHD-DCD) aged 7-17 years were preprocessed and motion was mitigated using ICA-AROMA. Emotion control, inhibition, and shifting were assessed as subdomains of BR. Functional connectivity (FC) maps were computed for ten limbic, prefrontal, parietal and temporal regions of interest and were investigated for associations with BR subdomains across all participants as well as for significant group differences.ResultsMultiple FC patterns were associated with BR across all participants. Some FC patterns were associated with multiple BR subdomains, while others were associated with only one. Differences in BR were found only between children with ADHD (i.e. ADHD and ADHD-DCD) and those without ADHD (i.e. TD and DCD). FC differences were also found between children with and without ADHD.ConclusionsOur results show dimensional associations between BR subdomain scores and whole-brain FC and highlight the potential of these associative patterns as brain-based signatures of BR in children with and without ADHD.
